Abstract

OBJECTIVE

We aimed to discuss the mechanism of occurrence and progression of epilepsy through analyzing the expression changes of UCA1 and NF-Kb in temporal hippocampus and UCA1 in peripheral blood in rats with epilepsy induced by lithium chloride-pilocarpine.

MATERIALS AND METHODS

The lithium chloride-pilocarpine-induced epilepsy rat model was established; 1, 7, 14, 30, and 60 d after status epilepticus were selected as the time points of research. The expression levels of UCA1 and NF-kB in the hippocampus of rats and UCA1 in peripheral blood were detected and analyzed using quantitative Real-time PCR (qRT-PCR). The differences and correlations between expression levels of UCA1 and NF-kB at each time point of research in experimental group and control group were analyzed statistically.

RESULTS

Results showed that mRNA expression levels of UCA1 and NF-kB in brain tissues in experimental group were higher than those in control group at each time point. The change trend of expression levels of UCA1 and NF-kB with time was consistent. The expression level of UCA1 in peripheral blood in experimental group at each time point was higher than that in control group, and mRNA expression level of UCA1 in peripheral blood in experimental group was positively correlated with that in brain tissue.

CONCLUSIONS

The expressions of UCA1 and NF-Kb are in the dynamic change in the formation of epilepsy, suggesting that UCA1 may participate in the pathogenesis of epilepsy, so as to provide a potentially feasible new direction for guiding the clinical diagnosis and treatment of epilepsy.

通过分析氯化锂-匹罗卡品致痫大鼠颞叶海马 UCA1 和 NF-Kb 表达变化及外周血 UCA1,探讨癫痫发生发展的机制。

材料与方法

建立氯化锂-匹罗卡品诱导的癫痫大鼠模型,分别在癫痫持续状态后 1、7、14、30、60 d 作为研究时间点,采用实时荧光定量聚合酶链反应(qRT-PCR)检测各组大鼠海马 UCA1 和 NF-kB 表达水平及外周血 UCA1 表达水平,分析实验组与对照组各时间点 UCA1 和 NF-kB 表达水平的差异及相关性。

结果

结果显示,实验组大鼠脑组织中 UCA1 和 NF-kB 的 mRNA 表达水平在各时间点均高于对照组,其表达水平随时间的变化趋势一致,实验组各时间点外周血 UCA1 表达水平均高于对照组,且实验组外周血 UCA1 表达水平与脑组织呈正相关。

结论

UCA1 和 NF-Kb 的表达在癫痫的形成过程中呈动态变化,提示 UCA1 可能参与癫痫的发病机制,为指导癫痫的临床诊断和治疗提供了一个潜在可行的新方向。
