To explore whether neurotrophin receptor-interacting MAGE homolog (NRAGE) is involved in the intestinal ischemia-reperfusion (I/R) and its effect on the apoptosis of intestinal epithelial cells and the expression of occludin protein.
The level of NRAGE protein after the rat small intestine I/R was detected by immunohistochemical (IHC) . The level of NRAGE protein and mRNA in IEC-6 cells after hypoxia and reoxygenation were tested by Western blot and RT-PCR respectively . The IEC-6 cells were divided into four groups, including overexpression by lentivirus infection (Lv- group), interference (sh- group), lentivirus control (Lv-control group), and normal control group without lentivirus infection (NC group). The apoptosis of IEC-6 cells after infection was analyzed by flow cytometry. The level of the tight junction protein occludin was detected by Western blot.
The expression of NRAGE were highly increased in intestinal mucosa epithelial cells after I/R (<0.01). The proteins and mRNA levels of NRAGE were increased after 6 h of hypoxia in IEC-6 cells. Compared with the Lv-control group, the early apoptosis rate was raised (<0.01) and the level of occludin was reduced (<0.01) in Lv- group; while the early apoptosis rate was reduced (<0.01) and the level of occludin was raised in sh- group(<0.001).
NRAGE may be involved in intestinal I/R and promote the apoptosis and decrease occludin expression of intestinal epithelial cells.
探讨神经营养因子受体相互作用MAGE同源物(NRAGE)是否参与肠道缺血再灌注(I/R)及其对肠上皮细胞凋亡和闭合蛋白表达的影响。
采用免疫组织化学(IHC)检测大鼠小肠I/R后NRAGE蛋白水平。分别采用蛋白质免疫印迹法(Western blot)和逆转录聚合酶链反应(RT-PCR)检测缺氧复氧后IEC-6细胞中NRAGE蛋白和mRNA水平。将IEC-6细胞分为四组,包括慢病毒感染过表达组(Lv-组)、干扰组(sh-组)、慢病毒对照组(Lv-control组)和未感染慢病毒的正常对照组(NC组)。采用流式细胞术分析感染后IEC-6细胞的凋亡情况。采用蛋白质免疫印迹法检测紧密连接蛋白闭合蛋白的水平。
I/R后肠黏膜上皮细胞中NRAGE表达显著升高(<0.01)。IEC-6细胞缺氧6小时后,NRAGE的蛋白和mRNA水平均升高。与Lv-control组相比,Lv-组早期凋亡率升高(<0.01),闭合蛋白水平降低(<0.01);而sh-组早期凋亡率降低(<0.01),闭合蛋白水平升高(<0.001)。
NRAGE可能参与肠道I/R,促进肠上皮细胞凋亡并降低闭合蛋白表达。
