To investigate dynamic contrast enhanced magnetic resonance imaging (DCE-MRI) for assessing histopathological and molecular biological features in induced rat epithelial ovarian carcinomas (EOCs).
7,12-dimethylbenz[A]anthracene (DMBA) was applied to induce EOCs in situ in 46 SD rats. Conventional MRI and DCE-MRI were performed to evaluate the morphology and perfusion features of the tumors, including the time-signal intensity curve (TIC), volume transfer constant (K), rate constant (K), extravascular extracellular space volume ratio (V) and initial area under the curve (IAUC). DCE-MRI parameters were correlated with histological grade, microvascular density (MVD), vascular endothelial growth factor (VEGF) and fraction of Ki67-positive cells and the serum level of cancer antigen 125 (CA125).
Thirty-five of the 46 rats developed EOCs. DCE-MRI showed type III TIC more frequently than type II (29/35 vs. 6/35, p < 0.001) in EOCs. The two types of TIC of tumors had significant differences in the histological grade, MVD, expression of VEGF and Ki67, and the serum level of CA125 (all p < 0.01). K, K and IAUC values showed significant differences in different histological grades in overall and pairwise comparisons except for IAUC in grade 2 vs. grade 3 (all p < 0.01). There was no significant difference in V values among the three grade groups (p > 0.05). K, K and IAUC values were positively correlated with MVD, VEGF and Ki67 expression (all p < 0.01). V was not significantly correlated with MVD, VEGF expression, Ki67 expression and the CA125 level (all p > 0.05).
TIC types and perfusion parameters of DCE-MRI can reflect tumor grade, angiogenesis and cell proliferation to some extent, thereby helping treatment planning and predicting prognosis.
探讨动态对比增强磁共振成像(DCE-MRI)在评估诱导性大鼠卵巢上皮性癌(EOC)的组织病理学和分子生物学特征中的作用。
采用 7,12-二甲基苯并蒽(DMBA)原位诱导 46 只 SD 大鼠发生 EOC。对肿瘤的形态学和灌注特征进行常规 MRI 和 DCE-MRI 检查,包括时间信号强度曲线(TIC)、容积转移常数(K)、速率常数(K)、血管外细胞外容积比(V)和初始曲线下面积(IAUC)。DCE-MRI 参数与组织学分级、微血管密度(MVD)、血管内皮生长因子(VEGF)、Ki67 阳性细胞比例以及血清癌抗原 125(CA125)水平相关。
46 只大鼠中 35 只发展为 EOC。DCE-MRI 显示 EOC 中 TIC 类型 III 比类型 II 更常见(29/35 比 6/35,p<0.001)。两种 TIC 类型的肿瘤在组织学分级、MVD、VEGF 和 Ki67 的表达以及 CA125 血清水平方面存在显著差异(均 p<0.01)。除 2 级与 3 级之间的 IAUC 外(均 p<0.01),在总体和两两比较中,不同组织学分级的 K、K 和 IAUC 值存在显著差异。三组间 V 值无显著差异(p>0.05)。K、K 和 IAUC 值与 MVD、VEGF 表达和 Ki67 表达呈正相关(均 p<0.01)。V 与 MVD、VEGF 表达、Ki67 表达和 CA125 水平无显著相关性(均 p>0.05)。
DCE-MRI 的 TIC 类型和灌注参数在一定程度上可以反映肿瘤分级、血管生成和细胞增殖,有助于治疗计划和预测预后。
