Abnormal expression of several microRNAs (miRNAs) has been demonstrated in many types of cancer tumor tissue. The miR-10b and miR-21 are an oncogenic miRNAs which play role in proliferation and invasion of Breast Cancer (BC) tumorigenesis. The aim of this study was to evaluate the miR-21 and miR-10b expression in BC in West Sumatran women, Indonesia.
A total of 40 samples, consisting of 30 samples of breast cancer tissues (BC) and 10 samples of fibroadenoma tissues (FATs) as control and non-cancerous were analyzed. The miR-21 and miR-10b expression of each sample were investigated by using realtime PCR, followed by universal Reverse Transcription (RT) then real-time PCR amplification with specific primers. Hsa-miR-16-5p LNA PCR primer was used as an endogenous control.
The results showed that the expression level of miR-21 was more than 4 times higher in BC than in FATs. The expression level of miR-10b was lower in BC than FATs, by a factor of 3.34 fold. Both these differences were statistically different (p = 0.001).
In this study it was concluded that for this sample of West Sumatran Women miR-21 expression in BC was higher than in FATs, whereas miR-10b was lower in BC than in FATs.
多种微小RNA(miRNA)的异常表达已在多种类型的癌症肿瘤组织中得到证实。miR-10b和miR-21是致癌性miRNA,在乳腺癌(BC)肿瘤发生的增殖和侵袭过程中发挥作用。本研究的目的是评估印度尼西亚西苏门答腊女性乳腺癌中miR-21和miR-10b的表达情况。
共分析40个样本,其中包括30个乳腺癌组织样本(BC)和10个纤维腺瘤组织样本(FATs)作为对照和非癌组织。通过实时PCR检测每个样本中miR-21和miR-10b的表达,先进行通用逆转录(RT),然后用特异性引物进行实时PCR扩增。使用Hsa-miR-16-5p LNA PCR引物作为内参。
结果显示,BC中miR-21的表达水平比FATs高4倍多。BC中miR-10b的表达水平比FATs低,低3.34倍。这两个差异均具有统计学意义(p = 0.001)。
在本研究中得出结论,对于西苏门答腊女性的这个样本,BC中miR-21的表达高于FATs,而BC中miR-10b的表达低于FATs。
