A Gram-stain-variable, strictly anaerobic, rod-shaped, catalase-negative and endospore-forming bacterial strain, designated MJC39, was isolated from grass silage preserved in Hokkaido, Japan. Growth occurred at 20-42 °C, pH 5.0-7.0 and NaCl concentrations up to 2 % (w/v). The isolated strain MJC39 produced butyric acid in peptone yeast extract medium with glucose. The DNA G+C content of strain MJC39 was 34.4±0.2 mol%. The major cellular fatty acids (>10 %) were C14 : 0, C16 : 0 and summed feature 3 (including C16 : 1ω7c/C16 : 1ω6c). No respiratory quinones were detected. The polar lipids of strain MJC39 were phosphatidylglycerol, phosphatidylethanolamine, diphosphatidylglycerol, one unidentified lipid, one unidentified aminolipid, two unidentified glycolipids, one unidentified phospholipid, one unidentified aminoglycolipid and one unidentified phosphoaminoglycolipid. Phylogenetic analysis based on 16S rRNA gene sequences showed that strain MJC39 was a member of the genus Clostridium and is closely related to Clostridium tyrobutyricum JCM 11008 (95.8 % similarity) and Clostridium algifaecis MB9-7 (95.5 % similarity). Based on the genotypic, phenotypic and chemotaxonomic characteristics, strain MJC39 represents a novel species of the genus Clostridium, for which the name Clostridium pabulibutyricum sp. nov. is proposed. The type strain is MJC39 (=JCM 31506=DSM 103944).
从日本北海道保存的青贮牧草中分离出一株革兰氏染色可变、严格厌氧、杆状、过氧化氢酶阴性且形成芽孢的细菌菌株,命名为MJC39。该菌株在20 - 42°C、pH 5.0 - 7.0以及NaCl浓度高达2%(w/v)的条件下生长。分离菌株MJC39在含有葡萄糖的蛋白胨酵母提取物培养基中产生丁酸。菌株MJC39的DNA G + C含量为34.4±0.2 mol%。主要细胞脂肪酸(>10%)为C14:0、C16:0和特征性总和3(包括C16:1ω7c/C16:1ω6c)。未检测到呼吸醌。菌株MJC39的极性脂质为磷脂酰甘油、磷脂酰乙醇胺、二磷脂酰甘油、一种未鉴定的脂质、一种未鉴定的氨基脂质、两种未鉴定的糖脂、一种未鉴定的磷脂、一种未鉴定的氨基糖脂和一种未鉴定的磷酸氨基糖脂。基于16S rRNA基因序列的系统发育分析表明,菌株MJC39是梭菌属的成员,与酪丁酸梭菌JCM 11008(相似度95.8%)和嗜藻梭菌MB9 - 7(相似度95.5%)密切相关。基于基因型、表型和化学分类学特征,菌株MJC39代表梭菌属的一个新物种,为此提出名称丁酸青贮梭菌(Clostridium pabulibutyricum sp. nov.)。模式菌株为MJC39(=JCM 31506 = DSM 103944)。
