To observe the effect of chiropractics on intervertebral disk extracell matrix metalloproteinase (MMP)-1, MMP-3, MMP-13, cellular collagen type Ⅱ (COL-Ⅱ), tissue inhibitor 1 of metalloproteinase (TIMP-1) and extracellular matrix (ECM) in the rats with cervical spondylosis, so as to explore its mechanism underlying improvement of cervical spondylosis.
A total of 40 SD rats were randomized into a sham group, a model group, a chiropractics group and a mobic group, 8 rats in each one. The model was established by static-dynamic imbalance method. The rats in the chiropractics group were treated with chiropractic therapy for two courses, 14 times a day and 14 days as a course. The rats in the mobic group were treated with intragastric administration of mobic. The expression of MMP-1, MMP-3, MMP-13, COL-Ⅱ and TIMP-1 in the intervertebral disk tissue were detected by immunohistochemistry, and the protein expression of ECM was detected by Western blot.
Compared with those in the sham group, the expressions of MMP-1, MMP-3, MMP-13 in the model group increased (all <0.05), and COL-Ⅱ, TIMP-1 decreased (both <0.05).Compared with those in the model group, the expressions of MMP-1, MMP-3, MMP-13 in the chiropractics group and the mobic group decreased (all <0.05), and COL-Ⅱ, TIMP-1 increased (all <0.05). Compared with that in the sham group, the expression of ECM protein in the model group decreased (<0.05). Compared with that in the model group, the expression of ECM protein in the chiropractics group and the mobic group increased (both <0.05).
Chiropractics may improve the degeneration in rats with cervical spondylosis by regulating intervertebral disc ECM system and relevant metabolic enzymes.
观察整脊疗法对颈椎病大鼠椎间盘细胞外基质金属蛋白酶(MMP)-1、MMP-3、MMP-13、Ⅱ型胶原(COL-Ⅱ)、金属蛋白酶组织抑制剂1(TIMP-1)及细胞外基质(ECM)的影响,以探讨其改善颈椎病的作用机制。
将40只SD大鼠随机分为假手术组、模型组、整脊疗法组和莫比可组,每组8只。采用动静力失衡法建立模型。整脊疗法组大鼠接受整脊治疗两个疗程,每天1次,每次14分钟,14天为一个疗程。莫比可组大鼠灌胃给予莫比可。采用免疫组织化学法检测椎间盘组织中MMP-1、MMP-3、MMP-13、COL-Ⅱ和TIMP-1的表达,采用蛋白质印迹法检测ECM的蛋白表达。
与假手术组相比,模型组MMP-1、MMP-3、MMP-13的表达升高(均P<0.05),COL-Ⅱ、TIMP-1的表达降低(均P<0.05)。与模型组相比,整脊疗法组和莫比可组MMP-1、MMP-3、MMP-13的表达降低(均P<0.05),COL-Ⅱ、TIMP-1的表达升高(均P<0.05)。与假手术组相比,模型组ECM蛋白表达降低(P<0.05)。与模型组相比,整脊疗法组和莫比可组ECM蛋白表达升高(均P<0.05)。
整脊疗法可能通过调节椎间盘ECM系统及相关代谢酶来改善颈椎病大鼠的退变。
