To investigate the detection of a human leukocyte antigen-B (HLA-B) allele by dual allele-specific real-time polymerase chain reaction (PCR) in patients with trichlorethylene-induced dermatitis. A total of 20 patients with trichlorethylene-induced dermatitis who were admitted and treated from January 2014 to October 2016 were enrolled as case group, and 20 persons who underwent physical examination from January to October, 2016 were enrolled as control group. Peripheral cubital venous blood samples were collected from all subjects, and dual allele-specific real-time PCR was used to detect the gene. The two groups were compared in terms of the proportion of subjects carrying gene. There were no significant differences between the case group and the control group in median age (25.0 years 27.0 years, =0.30, >0.05) and the proportion of male subjects (60.0% 70.0%, χ(2)=0.44, >0.05) . The mean time of exposure to trichloroethylene was 30.8 days in the case group, while the subjects in the control group were not exposed to trichloroethylene. The case group had a significantly higher frequency of gene than the control group (80.0% 20.0%, χ(2)=14.40, <0.01) with an odds ratio of 16.00. Dual allele-specific real-time PCR can be used for detection of the gene in patients with trichlorethylene-induced dermatitis.
探讨采用双等位基因特异性实时聚合酶链反应(PCR)检测三氯乙烯致皮炎患者的人类白细胞抗原B(HLA - B)等位基因。选取2014年1月至2016年10月收治的三氯乙烯致皮炎患者作为病例组,选取2016年1月至10月接受体检的人员作为对照组。采集所有受试者的外周肘静脉血样本,采用双等位基因特异性实时PCR检测该基因。比较两组携带该基因受试者的比例。病例组与对照组在年龄中位数(25.0岁对27.0岁,Z = 0.30,P>0.05)和男性受试者比例(60.0%对70.0%,χ² = 0.44,P>0.05)方面无显著差异。病例组三氯乙烯平均暴露时间为30.8天,而对照组受试者未接触三氯乙烯。病例组该基因频率显著高于对照组(80.0%对20.0%,χ² = 14.40,P<0.01),优势比为16.00。双等位基因特异性实时PCR可用于检测三氯乙烯致皮炎患者的该基因。
