To analyze the relationship among electrical conductivity (EC), total volatile basic nitrogen (TVB-N), which is an index of decomposition rate for meat production, and postmortem interval (PMI). To explore the feasibility of EC as an index of cadaveric skeletal muscle decomposition rate and lay the foundation for PMI estimation.
Healthy Sprague-Dawley rats were sacrificed by cervical vertebrae dislocation and kept at 28 ℃. Muscle of rear limbs was removed at different PMI, homogenized in deionized water and then skeletal extraction liquid of mass concentration 0.1 g/mL was prepared. EC and TVB-N of extraction liquid were separately determined. The correlation between EC (₁) and TVB-N (₂) was analyzed, and their regression function was established. The relationship between PMI () and these two parameters were studied, and their regression functions were separately established.
The change trends of EC and TVB-N of skeletal extraction liquid at different PMI were almost the same, and there was a linear positive correlation between them. The regression equation was ₂=0.14₁-164.91(²=0.982). EC and TVB-N of skeletal muscle changed significantly with PMI, and the regression functions were =19.38₁³-370.68₁²+2 526.03 ₁-717.06(²=0.994), and =2.56₂³-48.39₂²+330.60₂-255.04(²=0.997), respectively.
EC and TVB-N of rat postmortem skeletal muscle show similar change trends, which can be used as an index for decomposition rate of cadaveric skeletal muscle and provide a method for further study of late PMI estimation.
分析电导率(EC)、作为肉类生产分解率指标的总挥发性盐基氮(TVB-N)与死后间隔时间(PMI)之间的关系。探讨将EC作为尸体骨骼肌分解率指标的可行性,为PMI估计奠定基础。
通过颈椎脱臼法处死健康的Sprague-Dawley大鼠,并置于28℃环境。在不同的PMI时间点取出后肢肌肉,在去离子水中匀浆,然后制备质量浓度为0.1 g/mL的骨骼肌提取液。分别测定提取液的EC和TVB-N。分析EC(₁)与TVB-N(₂)之间的相关性,并建立其回归函数。研究PMI()与这两个参数之间的关系,并分别建立其回归函数。
不同PMI下骨骼肌提取液的EC和TVB-N变化趋势基本相同,且两者呈线性正相关。回归方程为₂=0.14₁-164.91(²=0.982)。骨骼肌的EC和TVB-N随PMI显著变化,回归函数分别为=19.38₁³-370.68₁²+2 526.03 ₁-717.06(²=0.994),以及=2.56₂³-48.39₂²+330.60₂-255.04(²=0.997)。
大鼠死后骨骼肌的EC和TVB-N呈现相似的变化趋势,可作为尸体骨骼肌分解率的指标,为后期PMI估计的进一步研究提供方法。
