Abstract

The main goal of this study was to investigate the possibility of expressing recombinant protein of C-C chemokine ligand 21 (CCL21) in via agroinfiltration. CCL21 is a chemokine can be used for anti-metastatic of cancer cell lines. To examine the expression of CCL21 protein in , the construct of was synthesized. This construct was cloned into pBI121 and the resulting CCL21 plasmid was agro-infiltrated into leaves. Within three days after infiltration, Expression of the foreign gene was confirmed by quantitative Real-time PCR. A recombinant CCL21 protein was immunogenically detected by western blot, dot blot and ELISA assay. And results showed that the foreign gene was expressed in the transformed leaves in high level. Also scratch assay was used to investigate the role of this protein in anti-metastatic function. The results demonstrated anti-metastatic of cancer cells in the presence of this protein.

摘要

本研究的主要目标是研究通过农杆菌浸润在[具体植物名称未给出]中表达重组C-C趋化因子配体21(CCL21)蛋白的可能性。CCL21是一种可用于癌细胞系抗转移的趋化因子。为检测CCL21蛋白在[具体植物名称未给出]中的表达,合成了[具体构建体名称未给出]的构建体。将该构建体克隆到pBI121中,所得的CCL21质粒通过农杆菌浸润导入[具体植物名称未给出]叶片。浸润后三天内,通过定量实时PCR确认外源基因的表达。通过蛋白质免疫印迹、斑点印迹和ELISA检测免疫原性重组CCL21蛋白。结果表明外源基因在转化叶片中高水平表达。还使用划痕试验研究该蛋白在抗转移功能中的作用。结果证明在该蛋白存在下癌细胞具有抗转移能力。
