To explore the analgesic effect of electroacupuncture(EA)by modulating 5-hydroxytryptamine 7 (5-HT) receptor in periaqueductal gray (PAG) and plasma calcitonin gene-related peptide (CGRP).
Forty-two male Sprague Dawley (SD) rats were randomly divided into control,model and EA groups, 14 rat in each one. The neurogenic migraine model was established by repeated electrical stimulation on sagittal sinus duramater. Intracranial electrodes were used in the control group without stimuli. The rats in the EA group received EA (0.5-1 mA, 2 Hz/15 Hz) at "Fengchi" (GB 20) for 10 min after dural electrical stimulation, once a day for 6 days. The expression of 5-HT receptor in the PAG was assessed by immunofluorescence and Western blot, respectively; plasma CGRP was measured by radioimmunoassay.
Compared with the control group, the positive neuron number and protein expression of 5-HT receptor in PAG and plasma CGRP increased after model establishment (all <0.001). The above mentioned indexes were reversed in the EA group compared with those in the model group (the positive neuron number and protein expression of 5-HT receptor, <0.01; plasma CGRP, <0.05).
EA at GB 20 can down-regulate the expression of 5-HT7 receptor in the PAG and reduce the content of plasma CGRP in the rats of migraine.
通过调节中脑导水管周围灰质(PAG)中5-羟色胺7(5-HT)受体及血浆降钙素基因相关肽(CGRP),探讨电针(EA)的镇痛作用。
42只雄性Sprague Dawley(SD)大鼠随机分为对照组、模型组和电针组,每组14只。通过反复电刺激矢状窦硬脑膜建立神经源性偏头痛模型。对照组使用颅内电极但不给予刺激。电针组在硬脑膜电刺激后于“风池”(GB 20)穴给予电针(0.5-1 mA,2 Hz/15 Hz),持续10分钟,每天1次,共6天。分别采用免疫荧光和蛋白质印迹法评估PAG中5-HT受体的表达;采用放射免疫分析法检测血浆CGRP。
与对照组比较,造模后PAG中5-HT受体阳性神经元数量、蛋白表达及血浆CGRP均升高(均P<0.001)。与模型组比较,电针组上述指标均有逆转(5-HT受体阳性神经元数量及蛋白表达,P<0.01;血浆CGRP,P<0.05)。
电针GB 20可下调偏头痛大鼠PAG中5-HT7受体的表达,降低血浆CGRP含量。
