Abstract

OBJECTIVE

To observe the effect of acupotomy therapy on mRNA expressions of Bcl-2, Bax , Caspase-3 in posterior cervical extensor muscles in cervical spondylosis rabbits, and explore its mechanisms for apoptosis of cervical muscles.

METHODS

New Zealand rabbits were randomly divided into normal, model, electroacupuncture (EA) and acupotomy groups, 6 in each group. The cervical spondylosis model was established by forced head-bowing for a long term. After model establishment, acupotomy was used at the trapezius muscle starting point and attachment site of sternocleidomastoid muscle, etc., once a week, total 3 times. EA was used at "Tianzhu" (BL 10), "Jingbailao" (EX-HN 15) and "Dazhu" (BL 11) for 3 weeks, 20 min a time, 3 times a week. Bcl-2, Bax, Caspase-3 mRNA expressions in posterior cervical extensor muscles were detected by real-time PCR.

RESULTS

There was no significant difference in the expression of Bcl-2 mRNA among all the groups (>0.05). Compared with the normal group, Bax mRNA increased in the model group (<0.01), and that in the acupotomy group was lower than that in the model group (<0.01). The ratio of Bcl-2/Bax mRNA in the model group decreased significantly compared with that in the normal group (<0.01); in comparison with the model group, the ratio in the acupotomy group increased (<0.01); and that in the acupotomy group was higher than the ratio in the EA group (<0.05). The Caspase-3 mRNA expression in the model group increased compared with that in the normal group (<0.05), and its expression in the acupotomy group decreased compared with those in the model and EA groups (<0.05).

CONCLUSIONS

Acupotomy therapy can regulate the mRNA expressions of Bax and Caspase-3, and retard apoptosis in posterior cervical extensor muscles, therefore the strained muscles are relieved, which may be one of its mechanisms for improving cervical spondylosis.

目的

观察针刀疗法对颈椎病兔颈后伸肌中Bcl-2、Bax、Caspase-3 mRNA表达的影响,探讨其对颈肌细胞凋亡的作用机制。

方法

将新西兰兔随机分为正常组、模型组、电针组和针刀组,每组6只。采用长期强迫低头法建立颈椎病模型。造模后,在斜方肌起点及胸锁乳突肌附着点等部位行针刀治疗,每周1次,共3次。电针组取“天柱”(BL 10)、“颈百劳”(EX-HN 15)、“大杼”(BL 11)穴,治疗3周,每次20分钟,每周3次。采用实时荧光定量PCR法检测颈后伸肌中Bcl-2、Bax、Caspase-3 mRNA的表达。

结果

各组Bcl-2 mRNA表达差异无统计学意义(>0.05)。与正常组比较,模型组Bax mRNA表达升高(<0.01),针刀组低于模型组(<0.01)。模型组Bcl-2/Bax mRNA比值较正常组显著降低(<0.01);与模型组比较,针刀组该比值升高(<0.01);针刀组该比值高于电针组(<0.05)。与正常组比较,模型组Caspase-3 mRNA表达升高(<0.05),针刀组Caspase-3 mRNA表达低于模型组和电针组(<0.05)。

结论

针刀疗法可调节Bax、Caspase-3 mRNA表达,抑制颈后伸肌细胞凋亡,从而缓解肌肉劳损,这可能是其改善颈椎病的作用机制之一。
