Abstract

Recent genome-wide association studies identified susceptibility loci for esophageal squamous cell carcinoma (ESCC), the most common histological type of esophageal cancer, in the phospholipase C ε-1 gene (). The aim of the present study was to investigate whether polymorphisms of were associated with the prognosis of ESCC patients in a high-incidence region of northern China. The rs2274223 A/G and rs11599672T/G single-nucleotide polymorphisms (SNPs) were genotyped by polymerase chain reaction-ligase detection reaction method in 207 ESCC patients with survival information. The mean age ± standard deviation of the 207 ESCC patients was 60.3±7.9 years. Sex, age, smoking status and family history of upper gastrointestinal cancer were not found to be associated with the survival time of ESCC patients. The mean survival time of rs2274223 SNP A/A, A/G and G/G genotype carriers were 42.9, 43.4 and 46.3 months, respectively; for rs11599672 SNP T/T, T/G and G/G genotype carriers the survival time were 42.8, 43.8 and 42.7 months, respectively. There was no significant difference in survival time among the ESCC patients with different genotypes of rs2274223 and rs11599672 SNPs. In conclusion, rs227423 and rs11599672 SNPs cannot be used as predictive markers for the survival of ESCC patients from a high-incidence region of northern China.

近期全基因组关联研究在磷脂酶Cε-1基因中确定了食管鳞状细胞癌(ESCC,食管癌最常见的组织学类型)的易感基因座。本研究旨在调查该基因的多态性是否与中国北方高发地区ESCC患者的预后相关。采用聚合酶链反应-连接检测反应方法,对207例有生存信息的ESCC患者进行rs2274223 A/G和rs11599672T/G单核苷酸多态性(SNP)基因分型。207例ESCC患者的平均年龄±标准差为60.3±7.9岁。未发现性别、年龄、吸烟状况及上消化道癌家族史与ESCC患者的生存时间相关。rs2274223 SNP的A/A、A/G和G/G基因型携带者的平均生存时间分别为42.9、43.4和46.3个月;rs11599672 SNP的T/T、T/G和G/G基因型携带者的生存时间分别为42.8、43.8和42.7个月。rs2274223和rs11599672 SNPs不同基因型的ESCC患者生存时间无显著差异。总之,rs227423和rs11599672 SNPs不能作为中国北方高发地区ESCC患者生存的预测标志物。
