Unilateral ureteral obstruction (UUO) alters the expression of renin-angiotensin system (RAS) components and angiotensin 1-7 (Ang 1-7) as a main arm of RAS is affected by UUO. The role of Mas receptor antagonist (A779) was examined in renal hemodynamic responses to Ang 1-7 in 3-day UUO and UUO removal (RUUO) in rats.
Forty-five male Wistar rats were randomly divided into three groups of sham operated, UUO, and RUUO, while each group was divided into two subgroups treated with vehicle or A779. Renal blood flow (RBF) and renal vascular resistance (RVR) responses to graded Ang 1-7 infusion were measured at controlled renal perfusion pressure.
Mean arterial pressure response to Ang 1-7 was increased in vehicle-treated subgroup significantly ( < 0.05) when compared with A779-treated subgroup. However, such observation was not seen in UUO and RUUO rats. The graded Ang 1-7 infusion increased RBF and decreased RVR significantly in vehicle-treated rats ( < 0.005). Furthermore, a significant difference was found between vehicle and A779-treated subgroups in sham, UUO, and RUUO groups ( < 0.005).
Ang 1-7 could alter the kidney hemodynamics responses in ureteral obstruction models.
单侧输尿管梗阻(UUO)会改变肾素-血管紧张素系统(RAS)组分的表达,且作为RAS主要分支的血管紧张素1-7(Ang 1-7)也会受UUO影响。本研究检测了Mas受体拮抗剂(A779)在大鼠输尿管梗阻3天及梗阻解除(RUUO)后对Ang 1-7引起的肾血流动力学反应中的作用。
45只雄性Wistar大鼠随机分为假手术组、UUO组和RUUO组,每组再分为接受溶媒或A779处理的两个亚组。在控制性肾灌注压条件下,测量分级输注Ang 1-7时的肾血流量(RBF)和肾血管阻力(RVR)反应。
与接受A779处理的亚组相比,接受溶媒处理的亚组对Ang 1-7的平均动脉压反应显著增加(<0.05)。然而,在UUO和RUUO大鼠中未观察到这种现象。分级输注Ang 1-7可使接受溶媒处理的大鼠的RBF显著增加,RVR显著降低(<0.005)。此外,在假手术组、UUO组和RUUO组中,接受溶媒处理和接受A779处理的亚组之间存在显著差异(<0.005)。
在输尿管梗阻模型中,Ang 1-7可改变肾脏血流动力学反应。
