Abstract

OBJECTIVES

This article describes the flavonoid composition of the flowers of Trollius europaeus and the method of isolation thereof and provides an attempt at investigating the antioxidant activity of the isolated flavonoids and the antityrosinase activity of the extracts from the investigated material.

METHODS

The compositional data were acquired by combining results of nuclear magnetic resonance, ultraviolet spectroscopy, electrospray ionisation-tandem mass spectrometry (ESI-MS/MS) analyses and those of an analysis of the products of acid hydrolysis of the compounds. The antioxidant activity of the extracts was studied using the 2,2'-diphenyl-1-picrylhydrazyl assay and the tyrosinase inhibitory activity - with the use of mushroom tyrosinase.

KEY FINDINGS

Ten flavonoid derivatives of luteolin and apigenin were isolated from the flowers of T. europaeus and identified. The investigation into the antioxidant activity revealed that orientin 2″-O-α-arabinopyranoside (4) and orientin 2″-O-β glucopyranoside (5) had a significant antioxidant effect.

CONCLUSIONS

The studies conducted led to the development of a method of isolating flavonoid, potentially antioxidant, compounds from T. europaeus. They allowed to determine which of the investigated flavonoids demonstrated significant antioxidant activity and could be used as natural antioxidants.

本文描述了毛茛科翠雀属植物欧洲翠雀花的花色苷组成及其分离方法,并对分离得到的花色苷的抗氧化活性和所研究材料提取物的酪氨酸酶抑制活性进行了研究。

方法

通过结合核磁共振、紫外光谱、电喷雾串联质谱(ESI-MS/MS)分析结果以及化合物酸水解产物的分析结果,获得组成数据。采用 2,2'-二苯基-1-苦基肼(DPPH)法测定提取物的抗氧化活性,采用蘑菇酪氨酸酶测定提取物的酪氨酸酶抑制活性。

从欧洲翠雀花的花中分离鉴定出 10 种木犀草素和芹菜素的黄酮类衍生物。抗氧化活性研究表明,木犀草苷 2″-O-α-阿拉伯糖苷(4)和木犀草苷 2″-O-β-吡喃葡萄糖苷(5)具有显著的抗氧化作用。

结论

本研究建立了一种从欧洲翠雀花中分离具有潜在抗氧化作用的黄酮类化合物的方法。确定了所研究的黄酮类化合物中哪些具有显著的抗氧化活性,可以用作天然抗氧化剂。
