Abstract

OBJECTIVE

To observe the effect of excretory/secretory products from adult worms (AES) on cecal ligation and puncture (CLP) -induced sepsis in mice.

METHODS

Forty-eight BALB/c mice were randomly divided into 3 groups:a sham operation group (PBS + sham group, Group A), a CLP-induced sepsis group (PBS+CLP group, Group B) and an AES treatment group (AES+CLP group, Group C). The mice of each group were intraperitoneally injected with 25 μg of AES or PBS only as a control in a total volume of 200 μl. Eight mice from each group were selected randomly for survival analysis of 96 hours. The other 8 mice in each group were observed for pathological changes in the lung, liver and kidney tissues by HE staining 12 h after CLP, and then determined for the detection of cytokines including TNF-α, IL-1β, IL-6, IL-10 and TGF-β in the sera by ELISA.

RESULTS

The difference among the survival rates of mice in the 3 groups was statistically significant ( = 21.16, < 0.05). Compared to Group A (100%), the survival rate of mice in Group B (0) decreased significantly ( < 0.05), and also the pathological damage degrees in the lung, liver and kidney tissues of the mice in Group B increased significantly after CLP. Compared with the mice in group B, the survival rate of those in Group C (70%) increased significantly ( < 0.05), and the pathological damage degrees in the lung, liver and kidney tissues of the mice in Group C decreased significantly after the treatment with AES. The differences among the levels of pro-inflammatory cytokines TNF-α ( = 27.11, < 0.05), IL1β ( = 18.75, < 0.05) and IL-6 ( = 100.93, < 0.05) in the sera of the mice in the three groups were statistically significant. Compared with the mice in Group A, the levels of the 3 cytokines of those in Group B increased significantly (all < 0.05). However, after the treatment with AES, the levels of the pro-inflammatory cytokines of those in Group C decreased significantly (all < 0.05). The differences among the levels of immunoregulatory cytokines IL-10 ( = 10.88, < 0.05) and TGF-β ( = 11.37, < 0.05) in the sera of the mice in the three groups were also statistically significant. Compared with the mice in Group B, the levels of IL-10 and TGF-β of those in Group C were higher after treatment with AES (both < 0.05).

CONCLUSIONS

AES has a therapeutic potential for alleviating sepsis induced by CLP in mice.

目的

观察成虫排泄/分泌产物(AES)对小鼠盲肠结扎穿刺(CLP)诱导的脓毒症的影响。

方法

48只BALB/c小鼠随机分为3组:假手术组(PBS+假手术组,A组)、CLP诱导的脓毒症组(PBS+CLP组,B组)和AES治疗组(AES+CLP组,C组)。每组小鼠腹腔注射25μg AES或仅注射PBS作为对照,总体积200μl。每组随机选取8只小鼠进行96小时的生存分析。每组另外8只小鼠在CLP术后12小时通过HE染色观察肺、肝和肾组织的病理变化,然后通过ELISA法测定血清中细胞因子TNF-α、IL-1β、IL-6、IL-10和TGF-β的含量。

结果

3组小鼠的生存率差异有统计学意义(=21.16,<0.05)。与A组(100%)相比,B组小鼠的生存率(0)显著降低(<0.05),且CLP术后B组小鼠肺、肝和肾组织的病理损伤程度明显增加。与B组小鼠相比,C组小鼠的生存率(70%)显著提高(<0.05),且AES治疗后C组小鼠肺、肝和肾组织的病理损伤程度明显降低。3组小鼠血清中促炎细胞因子TNF-α(=27.11,<0.05)、IL1β(=18.75,<0.05)和IL-6(=100.93,<0.05)水平的差异有统计学意义。与A组小鼠相比,B组小鼠的这3种细胞因子水平显著升高(均<0.05)。然而,AES治疗后,C组小鼠的促炎细胞因子水平显著降低(均<0.05)。3组小鼠血清中免疫调节细胞因子IL-10(=10.88,<0.05)和TGF-β(=11.37,<0.05)水平的差异也有统计学意义。与B组小鼠相比,AES治疗后C组小鼠的IL-10和TGF-β水平更高(均<0.05)。

结论

AES对减轻小鼠CLP诱导的脓毒症具有治疗潜力。
