Abstract

AsO-loaded nanoparticles (NPs) based on poly (lactic-co-glycolic acid) (PLGA) were prepared by double emulsion-solvent evaporation method. Then Lactose acid (LA) and/or PEG were modified onto the NPs by chemical covalent coupling through -NH and -COOH. The FTIR results showed that LA and PEG could be successfully modified onto the surface of the NPs. All the AsO-loaded PLGA NPs (AsO@PLGA NPs) presented suitable physical stability, favorable size, and spherical shape. The in vitro release rate of AsO from the NPs depended on the surface of the NPs. As expected, the AsO@PLGA-PEG/LA NPs showed a moderate release rate, the highest anticancer effects and cellular internalization against SMMC-7721 cell line. The PLGA-PEG/LA NPs could represent an effective nano-size delivery system of AsO for treatment of liver cancer.

载砷纳米粒子(NPs)基于聚(乳酸-共-乙醇酸)(PLGA)通过双重乳液-溶剂蒸发法制备。然后通过-NH 和-COOH 的化学共价偶联将乳糖酸(LA)和/或 PEG 修饰到 NPs 上。FTIR 结果表明,LA 和 PEG 可以成功修饰到 NPs 的表面上。所有载砷的 PLGA NPs(AsO@PLGA NPs)都表现出合适的物理稳定性、良好的尺寸和球形。砷从 NPs 中的体外释放率取决于 NPs 的表面。正如预期的那样,AsO@PLGA-PEG/LA NPs 表现出适中的释放率、对 SMMC-7721 细胞系的最高抗癌效果和细胞内化作用。PLGA-PEG/LA NPs 可以作为治疗肝癌的砷的有效纳米尺寸递药系统。
