[Effect of aerobic interval training on the expression of renal CD40 in a rat model with myocardial infarction and its mechanism].

Abstract

OBJECTIVE

To study the effects of aerobic interval training (AIT) on renal cluster of differentiation 40 (CD40) expression in rats with myocardial infarction (MI) and its possible mechanism.

METHODS

Thirty-six rats were randomly divided into three groups (=12):Sham, MI and MI with AIT (ME) groups. The MI model was established by ligation of the left anterior descending coronary artery. Treadmill training was performed five times a week for 8 weeks (AIT:60 min/day with 10 min of warm-up at 10 m/min and 50 min of exercise at 25 m/min 7 min interspersed with 3 min at 15 m/min). After training, cardiaorenal function and renal tissue remodeling were evaluated. The changes of CD40, high-sensitivity C reactive protein(hs-CRP), TNF-α, IL-6, p-NF-κBp65, blood urea nitrogen (BUN) and serum creatinine (sCr) were determined.

RESULTS

Compared with the sham group, MI significantly increased left ventricular end-diastolic pressure (LVEDP) and decreased left ventricular systolic pressure (LVSP) and left indoor pressure change rate peak (dp/dtmax) in the MI group, concomitant with the increase in renal collagen volume fraction (CVF), which was reversed by AIT in the ME group. Moreover, compared with the sham group, CD40 was largely dispersed within the cytoplasm of renal tubule cells in the MI group. Meanwhile, the expressions of renal CD40 mRNA and protein, the levels of serum and renal hs-CRP, TNF-α and IL-6, the phosphorylation of NF-κBp65 (p-NF-κBp65) and the levels of sCr and BUN were obviously increased in the MI group. Compared with the MI group, AIT decreased the expressions of renal CD40 mRNA and protein, the levels of serum and renal hs-CRP, TNF-α and IL-6 and the expression of p-NF-κBp65, as well as decreased the levels of sCr and BUN in the ME group.

CONCLUSIONS

AIT reduces the expressions of renal CD40 protein and mRNA, inhibits NF-κB signaling pathway, and then decreases the levels of inflammatory factors thereby improve the renal dysfunction after MI.

研究有氧间歇训练(AIT)对心肌梗死(MI)大鼠肾分化簇40(CD40)表达的影响及其可能机制。

方法

将36只大鼠随机分为三组(每组n = 12):假手术组、MI组和MI+AIT组。通过结扎左冠状动脉前降支建立MI模型。每周进行5次跑步机训练,持续8周(AIT组:每天60分钟,先以10米/分钟的速度热身10分钟,然后以25米/分钟的速度运动50分钟,期间穿插7分钟15米/分钟的运动和3分钟休息)。训练后,评估心肾功能和肾组织重塑情况。检测CD40、高敏C反应蛋白(hs-CRP)、肿瘤坏死因子-α(TNF-α)、白细胞介素-6(IL-6)、磷酸化核因子-κB p65(p-NF-κB p65)、血尿素氮(BUN)和血清肌酐(sCr)的变化。

结果

与假手术组相比,MI组左心室舒张末期压力(LVEDP)显著升高,左心室收缩压(LVSP)和左室内压变化率峰值(dp/dtmax)降低,同时肾胶原容积分数(CVF)增加,而MI+AIT组经AIT干预后上述指标得到改善。此外,与假手术组相比,MI组肾小管细胞胞质内CD40大量分散。同时,MI组肾CD40 mRNA和蛋白表达、血清和肾hs-CRP、TNF-α和IL-6水平、NF-κB p65磷酸化(p-NF-κB p65)以及sCr和BUN水平均明显升高。与MI组相比,MI+AIT组AIT降低了肾CD也0 mRNA和蛋白表达、血清和肾hs-CRP、TNF-α和IL-6水平以及p-NF-κB p65表达,同时降低了sCr和BUN水平。

结论

AIT降低肾CD40蛋白和mRNA表达,抑制NF-κB信号通路,进而降低炎症因子水平,从而改善MI后的肾功能障碍。
