The function of γ-amino butyric acid receptor (GR) was related with tinnitus. But, the effects of water extract (SP) on the mRNA expression of in mice with tinnitus were still unclear.
Eighteen SAMP8 mice were divided into the control group (intraperitoneal injection of saline, once per day), the tinnitus group (intraperitoneal injection of salicylate, 300 mg/kg body weight once per day), and the spirulina group [intraperitoneal injection of salicylate, 300 mg/kg body weight and oral SP supplementation (1000 mg/kg body weight) once per day]. Effects of SP on the mRNA expression of in the cochlea and brain of mice were studied for 4 days.
Compared to the control group, the tinnitus group had significantly higher tinnitus scores and lower mRNA expression of gene in the cochlear, brainstem, hippocampus and parahippocampus, temporal lobes, and the frontal lobes. On the other hand, the spirulina group had significantly lower tinnitus scores and higher gene expression than the tinnitus group in all tested areas.
SP could reduce salicylate-induced tinnitus possibly increasing the salicylate-induced down-regulation of gene expression.
γ-氨基丁酸受体(GR)的功能与耳鸣有关。但是,水提取物(SP)对耳鸣小鼠中[具体基因名称缺失]mRNA表达的影响仍不清楚。
将18只SAMP8小鼠分为对照组(腹腔注射生理盐水,每天一次)、耳鸣组(腹腔注射水杨酸盐,每天300mg/kg体重一次)和螺旋藻组[腹腔注射水杨酸盐,每天300mg/kg体重并口服SP补充剂(每天1000mg/kg体重)]。研究SP对小鼠耳蜗和脑中[具体基因名称缺失]mRNA表达的影响,持续4天。
与对照组相比,耳鸣组的耳鸣评分显著更高,而耳蜗、脑干、海马和海马旁回、颞叶以及额叶中[具体基因名称缺失]基因的mRNA表达更低。另一方面,在所有测试区域中,螺旋藻组的耳鸣评分显著低于耳鸣组,且[具体基因名称缺失]基因表达更高。
SP可能通过增加水杨酸盐诱导的[具体基因名称缺失]基因表达下调来减轻水杨酸盐诱导的耳鸣。
