Abstract

To investigate the effect of long non-coding RNA highly upregulated in liver cancer (lncRNA HULC) on the biological behavior of oral squamous cell carcinoma (OSCC) cell lines. A total of thirty patients with oral squamous cell carcinoma at Peking University Shenzhen Hospital from March 2017 to March 2018 were enrolled in this study. OSCC and adjacent tissues were extracted during tumor extensive resection. Quantitative real-time PCR (qPCR) was used to detect the expression levels of lncRNA HULC in OSCC and paracancerous tissues and OSCC cell lines. SCC15 and SCC25 cells were transfected with siRNA, and the effects of the gene on the biological behavior of OSCC cells were detected by cell counting assay, scratch assay, Transwell assay and Western blotting. The expression of lncRNA HULC in OSCC tissues (10.98±0.31, 30) was significantly higher than that in paracancerous tissues (8.39±0.31, 30) (5.93, 0.001), the expression of lncRNA HULC in OSCC cells (SCC15: 28.58±2.74; SCC25: 16.56±0.87; SCC9: 11.18±1.32; CAL27: 13.92±0.99, 5) was significantly higher than that in human keratinocytes (1.01±0.00, 5) ((SCC15)=10.08, (SCC25)=17.96, (SCC9)=7.71, (CAL27)13.09, 0.001). Down-regulation of lncRNA HULC in SCC15 and SCC25 cells can inhibit the proliferation, migration and invasion of tumor cells and promote tumor cell apoptosis. lncRNA HULC is highly expressed in OSCC and can enhance the proliferation, migration and invasion of cancer cells and inhibit tumor cell apoptosis.

摘要

研究肝癌中高表达的长链非编码RNA(lncRNA HULC)对口腔鳞状细胞癌(OSCC)细胞系生物学行为的影响。本研究纳入了2017年3月至2018年3月在北京大学深圳医院就诊的30例口腔鳞状细胞癌患者。在肿瘤广泛切除术中获取OSCC组织及其癌旁组织。采用定量实时聚合酶链反应(qPCR)检测lncRNA HULC在OSCC组织、癌旁组织及OSCC细胞系中的表达水平。用小干扰RNA(siRNA)转染SCC15和SCC25细胞,通过细胞计数实验、划痕实验、Transwell实验及蛋白质免疫印迹法检测该基因对OSCC细胞生物学行为的影响。lncRNA HULC在OSCC组织中的表达水平(10.98±0.31,n = 30)显著高于癌旁组织(8.39±0.31,n = 30)(t = 5.93,P = 0.001);lncRNA HULC在OSCC细胞系(SCC15:28.58±2.74;SCC25:16.56±0.87;SCC9:11.18±1.32;CAL27:13.92±0.99,n = 5)中的表达水平显著高于人角质形成细胞(1.01±0.00,n = 5)((SCC15)t = 10.08,(SCC25)t = 17.96,(SCC9)t = 7.71,(CAL27)t = 13.09,P = 0.001)。下调SCC15和SCC25细胞中lncRNA HULC的表达可抑制肿瘤细胞的增殖、迁移和侵袭,并促进肿瘤细胞凋亡。lncRNA HULC在OSCC中高表达,可增强癌细胞的增殖、迁移和侵袭能力,并抑制肿瘤细胞凋亡。
