Abstract

We developed -, -transfected human adipose stem cells (ASCs) to treat osteoarthritis (OA). -, -transfection led to chondrogenesis of adipose stem cells (ASCs) comparable to that achieved by growth factor treatment. Intra-articular injection of -, -transfected ASCs reduced the progression of surgically-induced OA in goats. tracking of transfected cell suggested paracrine mode of action. We think that -, -transfected ASC provides a novel potential strategy for the treatment of OA.

我们通过转染 -/- 基因的方法构建了人脂肪间充质干细胞(ASCs),以治疗骨关节炎(OA)。转染 -/- 基因能够诱导脂肪间充质干细胞(ASCs)向软骨细胞分化,其效果与生长因子治疗相当。关节内注射转染后的 ASCs 能够减缓山羊手术诱导型 OA 的进展。转染细胞的示踪表明其发挥作用的方式可能为旁分泌。我们认为转染 -/- 基因的 ASC 为 OA 的治疗提供了一种新的潜在策略。
