Natural Killer (NK) cells play a pivotal role in the immunosurveillance of Multiple Myeloma (MM), but it is still undefined whether the NK cell functional properties underlying their protective activity against MM are confined to distinct NK cell populations. Interestingly, herein we report that the CD56CD16 NK cell subset displayed higher cytolytic activity compared to the other NK cell subsets (i.e., CD56CD16, CD56CD16) against MM cells and its activity was impaired in MM patients. Decreased DNAM-1 expression levels were observed on the CD56CD16 NK cells during MM progression. Evaluating NK cell subset frequency after autologous hematopoietic stem cell transplantation, we found that CD56CD16 NK cells recovered earlier after transplantation. Overall, our data denote a key role of CD56CD16 subpopulation in the killing of MM cells and suggest that the reconstitution of CD56CD16 subpopulation after HSCT could be a useful approach of adoptive immunotherapy in the treatment of relapsed/refractory MM patients.
自然杀伤(NK)细胞在多发性骨髓瘤(MM)的免疫监视中起关键作用,但NK细胞针对MM的保护活性所基于的功能特性是否局限于不同的NK细胞群体仍不明确。有趣的是,在此我们报告,与其他NK细胞亚群(即CD56⁺CD16⁻、CD56⁻CD16⁺)相比,CD56⁻CD16⁺NK细胞亚群对MM细胞显示出更高的细胞溶解活性,且其活性在MM患者中受损。在MM进展过程中,CD56⁻CD16⁺NK细胞上观察到DNAX辅助分子-1(DNAM-1)表达水平降低。评估自体造血干细胞移植后NK细胞亚群频率,我们发现CD56⁻CD16⁺NK细胞在移植后恢复得更早。总体而言,我们的数据表明CD56⁻CD16⁺亚群在杀伤MM细胞中起关键作用,并提示HSCT后CD56⁻CD16⁺亚群的重建可能是治疗复发/难治性MM患者的一种有用的过继免疫疗法。
