The aim of this study was to determine the role of microencapsulated olfactory ensheathing cell (MC-OEC) transplantation in rats with sciatic nerve injury-induced pain, and its relationship with P2 × 7 receptor expression in the L4-5 spinal cord segment.
Olfactory bulb tissues of healthy Sprague Dawley (SD) rats were collected to culture olfactory ensheathing cells using differential attachment methods. Forty-eight healthy SD rats were randomly assigned to four groups: the sham, chronic constriction injury (CCI), olfactory ensheathing cell (OEC), and MC-OEC groups. Mechanical paw withdrawal thresholds were measured on days 7 and 14 after surgery. The expression of P2 × 7 receptor genes in the L4-5 spinal segment was detected by in situ hybridization and western blotting.
On post-surgical days 7 and 14, the mechanical paw withdrawal thresholds of rats in the MC-OEC, OEC and CCI were lower than that in the sham group. The mechanical paw withdrawal thresholds of rats in the MC-OEC and OEC were higher than that in the CCI group, compared with the OEC group. The mechanical paw withdrawal thresholds of rats in the MC-OEC were increased. The expression levels of P2 × 7 receptors in the L4-5 spinal cord segment in the CCI, OEC and MC-OEC were higher than that in the sham group, the expression levels of P2 × 7 receptors in the MC-OEC and OEC group were lower than that in the CCI group, compared with the OEC group, the expression levels of P2 × 7 receptors in the MC-OEC were decreased. All differences between groups were statistically significant (P value <0.05).
OEC and MC-OEC transplantation can relieve pain and reduce the expression level of P2 × 7 receptors in the L4-5 spinal cord segment. The therapeutic efficacy was better in the MC-OEC group than in the OEC group.
本研究旨在探讨微囊化嗅鞘细胞(MC-OEC)移植治疗大鼠坐骨神经损伤后痛觉的作用及其与 L4-5 脊髓节段 P2×7 受体表达的关系。
采用差速贴壁法培养健康 SD 大鼠嗅球组织中的嗅鞘细胞。将 48 只健康 SD 大鼠随机分为假手术组、慢性缩窄性损伤(CCI)组、嗅鞘细胞(OEC)组和微囊化嗅鞘细胞(MC-OEC)组。术后 7 天和 14 天测量机械性足底回缩阈值。采用原位杂交和 Western blot 检测 L4-5 脊髓节段 P2×7 受体基因的表达。
术后 7 天和 14 天,MC-OEC、OEC 和 CCI 组大鼠的机械性足底回缩阈值均低于假手术组,MC-OEC 和 OEC 组大鼠的机械性足底回缩阈值均高于 CCI 组,而 MC-OEC 组大鼠的机械性足底回缩阈值升高。CCI、OEC 和 MC-OEC 组大鼠 L4-5 脊髓节段 P2×7 受体的表达水平均高于假手术组,MC-OEC 和 OEC 组大鼠 L4-5 脊髓节段 P2×7 受体的表达水平均低于 CCI 组,而 MC-OEC 组大鼠 L4-5 脊髓节段 P2×7 受体的表达水平降低。组间差异均有统计学意义(P 值均<0.05)。
OEC 和 MC-OEC 移植均可缓解疼痛,降低 L4-5 脊髓节段 P2×7 受体的表达水平,MC-OEC 组的疗效优于 OEC 组。
