Abstract

Radix Cyathulae officinalis Kuan, one of the most well-known Chinese medicine, is widely used for invigorating livers and kidneys. The aim of this study is to investigate the hepatoprotective effects of polysaccharide from Radix Cyathulae officinalis Kuan (RCPS) against CCl-induced liver damage in rat. In vivo experiment showed that oral administrated of RCPS significantly decreased the production of ALT, AST, ALP, LDH, TNF-α, IL-6, IL-1β, MDA in serum, and reduced the expression of TLR4, P-iκBα, iκBα, NF-κBp65 in hepatic tissue. RCPS pretreatment also increased antioxidant enzyme activities Superoxide dismutase (SOD), Glutathione peroxidase (GSH-PX) and non-enzyme antioxidants glutathione (GSH), total antioxidant capacity (T-AOC) compared with CCl-induced. Furthermore, hepatic histopathological changes induced by CCl were significantly normalized by RCPS pretreatment. These results indicated that RCPS had hepatoprotective effect against CCl-induced acute liver injure.

摘要

怀牛膝是一种著名的中药,具有滋补肝肾的功效。本研究旨在探讨怀牛膝多糖(RCPS)对 CCl 诱导的大鼠肝损伤的保护作用。体内实验结果表明,RCPS 灌胃给药可显著降低 ALT、AST、ALP、LDH、TNF-α、IL-6、IL-1β、MDA 等血清酶和细胞因子的含量,降低 TLR4、P-iκBα、iκBα、NF-κBp65 等炎症相关蛋白的表达。RCPS 预处理还可提高 SOD、GSH-PX、GSH 等抗氧化酶和 T-AOC 的活性。此外,RCPS 预处理还可显著改善 CCl 诱导的肝组织病理损伤。综上所述,RCPS 对 CCl 诱导的急性肝损伤具有保护作用。
