Abstract

BACKGROUND

Breast cancer is a complex, heterogeneous disease and one of the most common malignancies in women worldwide. The efficacy of chemotherapy as an important breast cancer treatment option has been severely limited because of the inherent or acquired resistance of cancer cells. The molecular chaperone heat shock protein 90 (HSP90) upregulated in response to cellular stress is required for functions such as conformational maturation, activation and stability in more than 200 client proteins, mostly of the signaling type. In this study, the expression of HSP90 isoforms including HSP90α and HSP90 in breast cancer cell lines before and after treatment with doxorubicin (DOX) was assessed.

MATERIAL AND METHODS

The cell cytotoxicity of DOX in MDA-MB-231 and MCF-7 cell lines was determined using the MTT assay. Immunofluorescence and western blotting techniques were used to determine the expression of HSP90 in the cell lines before and after DOX treatment. Immunofluorescence was also conducted to ascertain the expression of HSP90α.

RESULTS

The MTT assay results showed that the MDA-MB- 231 cells (IC=14.521 μM) were more sensitive than the MCF-7 cells (IC=16.3315 μM) to DOX. The immunofluorescence results indicated that the expression of HSP90α in both cell lines decreased after exposure to DOX. The western blot and immunofluorescence analyses showed that HSP90 expression decreased in the MCF-7 cells but increased in the MDA-MB- 231 cells after DOX treatment. Conclusion: The obtained results suggested that HSP90α and HSP90 expression levels were reduced in the MCF-7 cells after exposure to DOX. In the MDA-MB-231 cells, HSP90α expression was reduced while HSP90 was found to be overexpressed following DOX treatment.

背景

乳腺癌是一种复杂的异质性疾病,也是全球女性中最常见的恶性肿瘤之一。由于癌细胞的固有耐药性或获得性耐药性,化疗作为一种重要的乳腺癌治疗选择,其疗效受到严重限制。分子伴侣热休克蛋白90(HSP90)在细胞应激反应中上调,是200多种客户蛋白(大多为信号类型)的构象成熟、激活和稳定性等功能所必需的。在本研究中,评估了阿霉素(DOX)处理前后乳腺癌细胞系中包括HSP90α和HSP90在内的HSP90亚型的表达。

材料与方法

采用MTT法测定DOX对MDA-MB-231和MCF-7细胞系的细胞毒性。利用免疫荧光和蛋白质印迹技术测定DOX处理前后细胞系中HSP90的表达。还进行了免疫荧光以确定HSP90α的表达。

结果

MTT法结果显示,MDA-MB-231细胞(IC = 14.521 μM)比MCF-7细胞(IC = 16.3315 μM)对DOX更敏感。免疫荧光结果表明,两种细胞系在暴露于DOX后HSP90α的表达均降低。蛋白质印迹和免疫荧光分析表明,DOX处理后,MCF-7细胞中HSP90表达降低,而MDA-MB-231细胞中HSP90表达增加。结论:所得结果表明,MCF-7细胞在暴露于DOX后HSP90α和HSP90表达水平降低。在MDA-MB-231细胞中,DOX处理后HSP90α表达降低,而HSP90被发现过表达。
