Abstract

BACKGROUND AND OBJECTIVES

is a bacterial sigma factor of RNA polymerase which is involved in the expression of the genes which control regulons and play a critical role in survival against stresses. Few suitable vectors are available which could be maintained successfully in cells and could in particular be used as a suicide vector to make mutation in the gene. The aim of this study was to investigate if mutagenesis has impact on bacterial morphology in addition to cell division.

MATERIALS AND METHODS

A 0.603 kb BamHI-PstI fragment subclone of pICRPOS38Ω was cloned into linearized pLYLO3. The final construct, pLRPOS38 suicide vector, was introduced into . Then the cytoplasm of mutant strain and wild-type were investigated by transmission electron microscopy.

RESULTS

After successful subcloning of suicide vector into , based on TEM study, it was demonstrated that mutation in gene leads to decomposition of outer membrane of .

CONCLUSION

A suitable vector to make suicide mutation in was constructed for .

背景与目的

是RNA聚合酶的一种细菌σ因子,参与控制调节子的基因表达,并在抵抗压力的生存中发挥关键作用。很少有合适的载体能够在 细胞中成功维持,特别是可以用作自杀载体在 基因中产生突变。本研究的目的是调查 诱变除了对细胞分裂有影响外,是否还会对细菌形态产生影响。

材料与方法
