OBJECTIVE
To observe the effect of electroacupuncture (EA) on expression of hypothalamic sirtuin 1(SIRT1) and proopiomelanocortin (POMC), and body weight, food-intake, blood glucose, and blood lipid levels in obese rats, so as to explore its mechanisms underlying improvement of obesity.
METHODS
Forty male Wistar rats were randomly divided into normal, model, EA, and sham EA groups (=10 rats in each group). The obesity model was established by feeding the rats with high fat diet. EA (2 Hz, 1 mA) was applied to "Zusanli" (ST36), "Zhongwan" (CV12), "Guanyuan" (CV4) and "Fenglong" (ST40) or sham acupoints (about 5 mm beside each acupoint, shallow needling) for 20 min, once every other day for 8 weeks. The rats' body weight and food-intake were recorded. The blood glucose (fasting plasma glucose: FPG, postprandial plasma glucose: PPG) and blood lipids (triglyceride: TG, total cholesterol: TC, non esterified fatty acid: NEFA) were assayed by using an automatic biochemical analyzer. The protein and mRNA expression levels of SIRT1 and POMC in the hypothalamus were detected by Western blot and quantitative real-time PCR, respectively.
RESULTS
In comparison with the normal group, the body weight, food-intake, blood lipids, and PPG levels were significantly increased (<0.05,<0.01), and the expression levels of SIRT1 protein and mRNA in the hypothalamus were significantly doun-regulated in the model group (<0.05). Following EA, the body weight, food-intake, blood lipids, and PPG levels were considerably down-regulated (<0.01,<0.05), and the expression levels of SIRT1 and POMC protein and mRNA in the hypothalamus were significantly up-regulated in the EA group rather than those in the sham EA and the model groups (<0.05)..
CONCLUSION
EA can reduce the obese rats' body weight, food-intake, blood lipids and blood glucose, which may be associated with its effect in up-regulating the SIRT1 and POMC expression of hypothalamus.
目的
观察电针(EA)对肥胖大鼠下丘脑沉默调节蛋白1(SIRT1)和阿黑皮素原(POMC)表达以及体重、进食量、血糖和血脂水平的影响,以探讨其改善肥胖的作用机制。
方法
将40只雄性Wistar大鼠随机分为正常组、模型组、电针组和假电针组(每组10只)。通过高脂饮食喂养建立肥胖模型。电针(2Hz,1mA)刺激“足三里”(ST36)、“中脘”(CV12)、“关元”(CV4)和“丰隆”(ST40)或假穴位(穴位旁约5mm,浅刺)20分钟,隔日1次,共8周。记录大鼠体重和进食量。使用自动生化分析仪检测血糖(空腹血糖:FPG,餐后血糖:PPG)和血脂(甘油三酯:TG,总胆固醇:TC,非酯化脂肪酸:NEFA)。分别采用蛋白质免疫印迹法和实时定量PCR检测下丘脑SIRT1和POMC的蛋白及mRNA表达水平。
结果
与正常组相比,模型组体重、进食量、血脂和PPG水平显著升高(P<0.05,P<0.01),下丘脑SIRT1蛋白和mRNA表达水平显著下调(P<0.05)。电针后,电针组体重、进食量、血脂和PPG水平显著下调(P<0.01,P<0.05),下丘脑SIRT1和POMC蛋白及mRNA表达水平显著上调,与假电针组和模型组相比差异有统计学意义(P<0.05)。
结论
电针可降低肥胖大鼠的体重、进食量、血脂和血糖,这可能与其上调下丘脑SIRT1和POMC表达的作用有关。
