The purpose of this study was to determine the optimum conditions for labeling genistein compounds with technetium-99m (Tc) radionuclides and the percentage of purity obtained in accordance with the requirements of the United State Pharmacopeia. The method used is optimization of several parameters including pH, SnCl.2HO as reducing agents, genistein concentration, and incubation time. The results showed that the optimum conditions for labeling Tc-Genistein were obtained under conditions of pH 8, the amount of SnCl.2HO reducing agents was 30 μg, 0.5 mg genistein, and in 10 min. The optimization of this condition resulted in radiochemical purity in the labeling of Tc-Genistein compounds at 95.43% ± 0.85%. The radiochemical purity of the labeling of Tc-Genistein compounds has met the requirements of the United State Pharmacopeia as a compound marked for diagnosis of more than 90%.
本研究的目的是确定用锝-99m(Tc)放射性核素标记染料木黄酮化合物的最佳条件以及根据美国药典要求获得的纯度百分比。所采用的方法是对几个参数进行优化,包括pH值、作为还原剂的SnCl₂·2H₂O、染料木黄酮浓度和孵育时间。结果表明,在pH值为8、SnCl₂·2H₂O还原剂用量为30μg、染料木黄酮0.5mg且孵育10分钟的条件下,获得了标记Tc-染料木黄酮的最佳条件。该条件的优化使得Tc-染料木黄酮化合物标记的放射化学纯度达到95.43%±0.85%。Tc-染料木黄酮化合物标记的放射化学纯度已符合美国药典对作为诊断用化合物超过90%的要求。
