Abstract

Sortase A (SrtA), a type of transpeptidase responsible for anchoring surface proteins to the peptidoglycan cell wall, is important in the virulence of gram-positive bacteria. Three compounds were isolated from marine-derived sp. MBTH32 using various chromatography techniques. The structures of these compounds were determined based on spectroscopic data and comparisons with previously reported data. Among the metabolites tested, lumichrome showed strong inhibitory activity against SrtA without affecting cell viability. The results of cell clumping activity assessment suggest the potential for using this compound to treat infection by inhibiting SrtA activity.

表面蛋白锚定于肽聚糖细胞壁的转肽酶 Sortase A(SrtA)在革兰氏阳性菌的毒力中起重要作用。三种化合物从海洋来源的 sp. MBTH32 中使用各种色谱技术分离得到。根据光谱数据和与先前报道的数据进行比较,确定了这些化合物的结构。在所测试的代谢物中,色胺酮对 SrtA 表现出强烈的抑制活性而不影响细胞活力。细胞聚集活性评估的结果表明,通过抑制 SrtA 活性,该化合物具有治疗 感染的潜力。
