To establish a method for simultaneous determination of L-carnitine, choline, taurin in infant by ultrafiltration tube cleaning and ultraperformance liquid chromatography-tandem mass spectrometry(UPLC-MS/MS).
The sample was dissolved in water and acid hydrolyzed, then adjusted pH with a sodium hydroxide solution. After ultrafiltration tube cleaning and centrifugation, the sample solution was separated on an amide column ACQUITY UPLC BEH Amide(2. 1 mm × 100 mm, 1. 7 μm) and detected by MS/MS under multiple reaction monitoring(MRM) mode. The quantification was performed by the internal standard calibration method.
The linear ranges were 1. 00-500 μg/L for L-carnitine and choline, 10-1000 μg/L for taurine with correlation coefficients of 0. 999. The mean recoveries were 89. 7%-107. 4% with relative standard deviations(RSD, n = 6) were2. 6%-8. 1%. The detection limits of the method were 0. 812 mg/kg for choline, 0. 623 mg/kg for L-carnitine and 9. 34 mg/kg for taurine, respectively.
This method is simple, accurate, reproducible and sensitivity.
建立一种采用超滤管净化结合超高效液相色谱-串联质谱法(UPLC-MS/MS)同时测定婴幼儿体内左旋肉碱、胆碱、牛磺酸的方法。
样品用水溶解并进行酸水解,然后用氢氧化钠溶液调节pH值。经超滤管净化和离心后,样品溶液在酰胺柱ACQUITY UPLC BEH Amide(2.1 mm×100 mm,1.7 μm)上进行分离,并在多反应监测(MRM)模式下通过MS/MS检测。采用内标校准法进行定量分析。
左旋肉碱和胆碱的线性范围为1.00 - 500 μg/L,牛磺酸的线性范围为10 - 1000 μg/L,相关系数均为0.999。平均回收率为89.7% - 107.4%,相对标准偏差(RSD,n = 6)为2.6% - 8.1%。该方法的检测限分别为胆碱0.812 mg/kg、左旋肉碱0.623 mg/kg、牛磺酸9.34 mg/kg。
该方法简便、准确、重现性好且灵敏度高。
