To study the mechanism and clinical significance of B-cell lymphoma/leukemia 11A (BCL11A) gene in the development of endometrial carcinoma (EC).
100 EC, 20 normal endometrium and 20 atypical hyperplasia endometrium specimens were collected from West China Second University Hospital from January 2005 to January 2015. The expression of BCL11A was detected by immunohistochemistry and its relationships with clinicopathological features and survival were analyzed.
The expression of BCL11A in ER-/PR-EC was higher than that in normal endometrium, atypical hyperplasia endometrium, and ER+/PR+EC (P < 0.001). The expression of BCL11A in EC was associated with age, menopause, EC classification, para-aortic lymph node metastasis, tumor differentiation, histological type, ER/PR expression and p53 expression (P < 0.05). The expression of BCL11A in the deceased group was significantly higher than that in the survival group (P = 0.011). Survival analysis indicated that the high expression of BCL11A was associated with low survival rate (P < 0.001).
BCL11A may play an important role in the development of ER-/PR-EC. It may serve as a potential target for therapy and a predict factor for prognosis. It can also provide molecular basis for new EC classification.
研究B细胞淋巴瘤/白血病11A(BCL11A)基因在子宫内膜癌(EC)发生发展中的机制及临床意义。
收集2005年1月至2015年1月四川大学华西第二医院的100例子宫内膜癌、20例正常子宫内膜及20例不典型增生子宫内膜标本。采用免疫组织化学法检测BCL11A的表达,并分析其与临床病理特征及生存情况的关系。
BCL11A在雌激素受体/孕激素受体阴性(ER-/PR-)的子宫内膜癌中的表达高于正常子宫内膜、不典型增生子宫内膜及雌激素受体/孕激素受体阳性(ER+/PR+)的子宫内膜癌(P<0.001)。子宫内膜癌中BCL11A的表达与年龄、绝经情况、子宫内膜癌分类、腹主动脉旁淋巴结转移、肿瘤分化程度、组织学类型、ER/PR表达及p53表达相关(P<0.05)。死亡组BCL11A的表达明显高于生存组(P=0.011)。生存分析表明,BCL11A高表达与低生存率相关(P<0.001)。
BCL11A可能在ER-/PR-的子宫内膜癌发生发展中起重要作用。它可能成为潜在的治疗靶点及预后预测因子。也可为子宫内膜癌新的分类提供分子依据。
