Abstract

(PG) is a widely used mushroom for health promotion. We studied the hepatoprotective effect of the polysaccharide aqueous extract of PG (PGP) against carbon tetrachloride (CCl)-induced liver injury in rats. Sprague Dawley rats were divided into 5 groups: Normal control, CCl control, PGP 50, 100, and 200 mg/kg + CCl. The levels of serum biochemical parameters, liver lipid peroxide and antioxidant enzymes, and histological appearances were evaluated. The CCl-induced increments of alanine aminotransferase, asparate aminotransferase, and alkaline phosphatase levels in serum were significantly decreased by PGP-pretreatments. The PGP dose-dependently decreased hepatic malondialdehyde formations in CCl-treatment groups. Hepatic antioxidant enzymes (superoxide dismutase, catalase, and glutathione peroxidase) were elevated by PGP in CCl-treatment groups. Histopathological evaluation of liver showed that the loss of hepatocytes, fatty changes, swelling and extensive necrosis of hepatocytes in centrilobular regions of the CCl-treated rats were ameliorated by PGP pretreatment. The PGP has hepatoprotective and antioxidative effects in CCl-induced liver injury of rat.

平菇(PG)是一种广泛用于促进健康的蘑菇。我们研究了平菇多糖水提取物(PGP)对四氯化碳(CCl)诱导的大鼠肝损伤的保肝作用。将Sprague Dawley大鼠分为5组:正常对照组、CCl对照组、PGP 50、100和200mg/kg + CCl组。评估了血清生化参数、肝脏脂质过氧化物和抗氧化酶水平以及组织学表现。PGP预处理显著降低了CCl诱导的血清中丙氨酸氨基转移酶、天冬氨酸氨基转移酶和碱性磷酸酶水平的升高。PGP剂量依赖性地降低了CCl处理组肝脏丙二醛的形成。PGP使CCl处理组的肝脏抗氧化酶(超氧化物歧化酶、过氧化氢酶和谷胱甘肽过氧化物酶)升高。肝脏组织病理学评估表明,PGP预处理改善了CCl处理大鼠小叶中心区域肝细胞的丢失、脂肪变性、肿胀和广泛坏死。PGP对CCl诱导的大鼠肝损伤具有保肝和抗氧化作用。
