The Biopharmaceutics Classification System (BCS) categorizes active pharmaceutical ingredients according to their solubility and permeability properties, which are susceptible to matrix or formulation effects. The aim of this research was to evaluate the matrix effects of a hydroethanolic extract of calyces from Physalis peruviana L. (HEE) and its butanol fraction (BF), on the biopharmaceutics classification of their major compound, quercetin-3-O-rutinoside (rutin, RU).
Rutin was quantified by HPLC-UV, and Caco-2 cell monolayer transport studies were performed to obtain the apparent permeability values (P ). Aqueous solubility was determined at pH 6.8 and 7.4.
The P values followed this order: BF > HEE > RU (1.77 ± 0.02 > 1.53 ± 0.07 > 0.90 ± 0.03 × 10 cm/s). The lowest solubility values followed this order: HEE > RU > BF (2.988 ± 0.07 > 0.205 ± 0.002 > 0.189 ± 0.005 mg/ml).
According to these results, rutin could be classified as BCS classes III (high solubility/low permeability) and IV (low solubility/low permeability), depending on the plant matrix. Further work needs to be done in order to establish how apply the BCS for research and development of new botanical drugs or for bioequivalence purposes.
生物药剂学分类系统(BCS)根据活性药物成分的溶解性和渗透性对其进行分类,而这些特性易受基质或制剂影响。本研究的目的是评估灯笼草萼片的乙醇提取物(HEE)及其正丁醇部位(BF)对其主要化合物槲皮素-3-O-芸香糖苷(芦丁,RU)生物药剂学分类的基质效应。
采用高效液相色谱-紫外检测法对芦丁进行定量,并进行Caco-2细胞单层转运研究以获得表观渗透率值(P)。在pH 6.8和7.4条件下测定水溶性。
P值遵循以下顺序:BF>HEE>RU(1.77±0.02>1.53±0.07>0.90±0.03×10⁻⁶ cm/s)。最低溶解度值遵循以下顺序:HEE>RU>BF(2.988±0.07>0.205±0.002>0.189±0.005 mg/ml)。
根据这些结果,芦丁可根据植物基质被分类为BCS III类(高溶解性/低渗透性)和IV类(低溶解性/低渗透性)。为了确定如何将BCS应用于新植物药的研发或生物等效性研究,还需要进一步开展工作。
