In this study, novel core-shell crosslinked dextran-hemin micelles were first successfully synthesized. Hemin grafted to dextran is not only used as a phototriggered switch for the controlled release of loaded 5-fluorouracil (5FU) quickly, but also simultaneously as a photosensitizer for photodynamic therapy under laser irradiation. The prepared micelles exhibit high stability against extensive dilution of solution which can restrain premature drug loss and reduce side effects to human bodies. The micelles possess good biocompatibility as well. More importantly, 5FU-loaded crosslinked dextran-hemin micelles present greater antitumor effect than free 5FU or crosslinked micelles under laser irradiation, indicating the excellent synergetic antitumor effect of hemin with 5FU in the as-prepared micelles. We are convinced that these phototriggered crosslinked micelles combining chemotherapy and photodynamic therapy have tremendous potential for localized cancer treatment.
在本研究中,首次成功合成了新型核壳交联葡聚糖-血红素胶束。接枝到葡聚糖上的血红素不仅用作光触发开关,用于快速控制释放负载的5-氟尿嘧啶(5FU),还同时用作激光照射下光动力疗法的光敏剂。所制备的胶束在溶液大量稀释时表现出高稳定性,可抑制药物过早流失并减少对人体的副作用。这些胶束还具有良好的生物相容性。更重要的是,负载5FU的交联葡聚糖-血红素胶束在激光照射下比游离5FU或交联胶束具有更强的抗肿瘤作用,表明所制备胶束中血红素与5FU具有优异的协同抗肿瘤作用。我们相信,这些结合化疗和光动力疗法的光触发交联胶束在局部癌症治疗方面具有巨大潜力。
