Abstract

OBJECTIVE

To detect the expressions of vitamin D receptor (VDR) in peripheral blood monocytes (PBMCs) and its association with monocyte chemoattractant protein-1 (MCP-1) in patients with systemic lupus erythemaotsus (SLE).

METHODS

We examined the expressions of VDR and MCP-1 mRNAs in the PBMCs in 60 SLE patients and 28 healthy individuals using real- time quantitative PCR. We also detected the expression of VDR protein in the PBMCs using Western blotting and peripheral blood MCP-1 level using ELISA for these participants. The correlation of VDR and MCP-1 expressions with the disease activity index of SLE (SLEDAI) of the patients were analyzed.

RESULTS

The expressions of VDR mRNA and protein in the PBMCs were significantly lower in patients with SLE than in the healthy individuals ( < 0.01), and that in the patients with active disease was lower than in remission ( < 0.05). The MCP-1 mRNA expression in the PBMCs and its serum levels were significantly increased in SLE patients as compared with the healthy individuals ( < 0.01), and the increase was significantly more obvious in the patients with active disease than in those in remission ( < 0.01). Pearson correlation analysis showed that VDR mRNA in the PBMCs was negatively correlated with SLEDAI (=-0.417, =0.001); negative correlations were also found between VDR mRNA and MCP-1 mRNA(=-0.554, =0.000) and between VDR protein expression and serum MCP-1 level (=-0.400, =0.028).

CONCLUSIONS

The down-regulation of VDR expression in the PBMC is negatively correlated with the disease activity of SLE. VDR may play an important role in the pathogenesis of SLE by affecting the expression of MCP-1.

目的

检测系统性红斑狼疮(SLE)患者外周血单核细胞(PBMCs)中维生素D受体(VDR)的表达及其与单核细胞趋化蛋白-1(MCP-1)的相关性。

方法

采用实时定量PCR检测60例SLE患者和28例健康个体PBMCs中VDR和MCP-1 mRNA的表达。同时采用蛋白质免疫印迹法检测这些参与者PBMCs中VDR蛋白的表达,采用酶联免疫吸附测定法检测外周血MCP-1水平。分析VDR和MCP-1表达与患者SLE疾病活动指数(SLEDAI)的相关性。

结果

SLE患者PBMCs中VDR mRNA和蛋白的表达显著低于健康个体(<0.01),且活动期患者低于缓解期患者(<0.05)。与健康个体相比,SLE患者PBMCs中MCP-1 mRNA表达及其血清水平显著升高(<0.01),且活动期患者的升高明显高于缓解期患者(<0.01)。Pearson相关性分析显示,PBMCs中VDR mRNA与SLEDAI呈负相关(r=-0.417,P=0.001);VDR mRNA与MCP-1 mRNA之间(r=-0.554,P=0.000)以及VDR蛋白表达与血清MCP-1水平之间(r=-0.400,P=0.028)也呈负相关。

结论
