The ploidy of a mutant of Saccharomyces cerevisiae defective in recombination (Rec-) has been determined using tetrad analysis and flowing fluorometry. Evidence is obtained that the effect of the Rec- phenotype, i.e. the increase of the stability of plasmids with 2 mkm DNA ori replication in the yeast cirO cells is not the result of the diploidy in cells of the Rec- mutant developed in the process of transformation.
利用四分体分析和流式荧光测定法确定了酿酒酵母中重组缺陷型(Rec-)突变体的倍性。有证据表明,Rec-表型的作用,即在酵母cirO细胞中具有2μm DNA ori复制的质粒稳定性增加,并不是转化过程中产生的Rec-突变体细胞的二倍体状态导致的。
