BACKGROUND/AIM: Interactions between colorectal cancer (CRC) cells and myofibroblasts govern many processes such as cell growth, migration, invasion and differentiation, and contribute to CRC progression. Robust experimental tests are needed to investigate the nature of these interactions for future anticancer studies. The purpose of the study was to design and validate in vitro assays for studying the communication between myofibroblasts and CRC epithelial cell lines.
The influence of co-culture of myofibroblasts and CRC cell lines is discussed using various in vitro assays including direct co-culture, transwell assays, Matrigel-based differentiation and cell invasion experiments.
The results from these in vitro assays clearly demonstrated various aspects of the crosstalk between myofibroblasts and CRC cell lines, which include cell growth, differentiation, migration and invasion.
The reported in vitro assays provide a basis for investigating the factors that control the myofibroblast-epithelial cell interactions in CRC in vivo.
背景/目的:结直肠癌(CRC)细胞与肌成纤维细胞之间的相互作用调控着细胞生长、迁移、侵袭和分化等多种过程,促进了结直肠癌的进展。为了进行未来的抗癌研究,需要进行强有力的实验来研究这些相互作用的本质。本研究的目的是设计并验证体外检测方法,以研究肌成纤维细胞与 CRC 上皮细胞系之间的通讯。
使用各种体外检测方法,包括直接共培养、Transwell 检测、基于 Matrigel 的分化和细胞侵袭实验,讨论了肌成纤维细胞与 CRC 细胞系共培养的影响。
这些体外检测的结果清楚地显示了肌成纤维细胞与 CRC 细胞系之间的串扰的各个方面,包括细胞生长、分化、迁移和侵袭。
