Abstract

A two-stage enzymatic hydrolysis method, in which phospholipase A (PLA) was added after phospholipase A (PLA) was added for a certain time, was successfully carried out to prepare glycerylphosphorylcholine (GPC) from soybean concentrated phospholipid. Effects of reaction variables on hydrolysis reaction were optimized using response surface methodology, and the optimal conditions were as follows: PLA load of 1.25%, PLA load of 0.70%, substrate concentration of 13%, reaction temperature of 41°C, and stirring rate of 680 rpm. Under the optimal conditions, the GPC yield reached 83.07%, which is close to the predicted value by the fitted model. This paper not only provides an efficient and low-cost method to prepare GPC, but also improves the high-value utilization of soybean concentrated phospholipid.

摘要

采用两步酶解方法,即在一定时间后添加磷脂酶 A(PLA),成功地从大豆浓缩磷脂中制备了甘油磷酸胆碱(GPC)。利用响应面法优化了水解反应的反应变量,优化条件如下:PLA 用量 1.25%、PLA 用量 0.70%、底物浓度 13%、反应温度 41°C、搅拌速率 680rpm。在最佳条件下,GPC 的产率达到 83.07%,接近拟合模型预测值。本研究不仅提供了一种高效、低成本的制备 GPC 的方法,而且提高了大豆浓缩磷脂的高值化利用。
