Abstract

Ischemic stroke is one of the major diseases with high morbidity, mortality, and disability rate all over the world. Chinese herb-derived active components would provide valuable candidate compounds for ischemic stroke therapy. Paeoniflorin (PF) is an active ingredient from Paeoniae Radix which possesses neurovascular effect after ischemia. However, so far, few studies are reported on the efficacy and mechanism of PF from angiogenesis aspects. Results from our studies showed that the ability for proliferation, migration, and tube formation in bone marrow-derived endothelial progenitor cells (BM-EPCs) was promoted by coculturing with PF (100 [Formula: see text]M). Furthermore, to investigate the angiogenic effects of PF , we constructed an ischemic stroke model in rats and found that PF could reduce cerebral infarction, alleviate pathological injury, and increase the secretion of pro-angiogenic factors and cerebral vascular density after intraperitonially administration of 40 mg ⋅ kg[Formula: see text] ⋅ day[Formula: see text] for 14 days. Up-regulating the expression of VEGF/VEGF-R might be the mechanism of PF's angiogenic action. In conclusion, the present study provides evidence that PF is an active monomer of Traditional Chinese Medicine which shows angiogenic actions on endothelial progenitor cells and in ischemic stroke rat model.

缺血性脑卒中是全世界发病率、死亡率和致残率都很高的主要疾病之一。中药来源的活性成分可为缺血性脑卒中治疗提供有价值的候选化合物。芍药苷(PF)是芍药根中的一种活性成分,具有缺血后神经血管作用。然而,到目前为止,很少有研究报道 PF 从血管生成方面的疗效和机制。我们的研究结果表明,PF(100 [Formula: see text]M)与骨髓来源的内皮祖细胞(BM-EPC)共培养可促进其增殖、迁移和管状形成能力。此外,为了研究 PF 的血管生成作用,我们构建了大鼠缺血性脑卒中模型,发现 PF 腹腔内给药 40 mg ⋅ kg[Formula: see text] ⋅ day[Formula: see text],连续 14 天,可减少脑梗死,减轻病理损伤,增加促血管生成因子的分泌和脑血管密度。上调 VEGF/VEGF-R 的表达可能是 PF 血管生成作用的机制。总之,本研究为 PF 作为中药的一种活性单体,对内皮祖细胞和缺血性脑卒中大鼠模型具有血管生成作用提供了证据。
