This study aimed to explore a set of simplified schemes for the preparation and application of artificial cerebrospinal fluid (ACSF) to improve the experiment efficiency and neurosafety of ACSF. We prepared ACSF into parts A and B, according to the cerebrospinal fluid (CSF) data in rabbits. They were mixed in equal volumes to form ACSF, continous foaming with mixture gas (95% O and 5% CO). Sampling inspection showed the chemical stability of ACSF in the three months after preparation. However, it needed to be kept continous foaming, as pH is correlated to the solubility of CO. We further improved the application scheme by sealing the foamed ACSF in infusion bags filled with mixture gas, which could keep the pH stable for 24 hours. It was helpful in promoting the progress of clinical and experimental research relating to ACSF.
本研究旨在探索一套简化的人工脑脊液(ACSF)制备和应用方案,以提高 ACSF 的实验效率和神经安全性。我们根据兔 CSF 数据,将 ACSF 制备成 A、B 两部分,按等体积混合形成 ACSF,用混合气体(95% O 和 5% CO)连续发泡。抽样检查表明,制备后三个月内 ACSF 的化学稳定性良好。但需要保持连续发泡,因为 pH 值与 CO 的溶解度有关。我们进一步改进了应用方案,将发泡的 ACSF 密封在充满混合气体的输液袋中,可使 pH 值稳定 24 小时。这有助于推动与 ACSF 相关的临床和实验研究的进展。
