Abstract

We report sequential metal catalysis towards indolocarbazole glycosides. The signature event is highlighted by i) Pd -catalyzed addition of indolocarbazole to alkoxyallene combined with ring-closing-metathesis; ii) Ru-catalyzed chemoselective olefin migration; iii) Pd -catalyzed oxidative cyclization to build the bicyclic core structure of the target compounds. This approach gave access to both natural pyranose- and non-natural septanose glycosides. A short formal synthesis of 7-oxostaurosporine was achieved via this strategy.

我们报告了吲哚咔唑糖苷的连续金属催化反应。标志性反应包括:i)Pd 催化吲哚咔唑与烷氧基丙二烯的加成,以及环 closing-metathesis;ii)Ru 催化的烯烃迁移的选择性反应;iii)Pd 催化的氧化环化反应,构建目标化合物的双环核心结构。这种方法可以得到天然吡喃糖和非天然庚糖糖苷。通过这种策略,实现了 7-氧代牛磺菌素的短形式合成。
