Abstract

Gagnep is a Vietnamese medicinal plant which has been commonly used in traditional and folk medicines for the treatment of different diseases. The goals of the present study are to investigate chemical composition and anti-proliferative activity of rhizome extract (CSE). The cytotoxicity of CSE was evaluated using WST-1 and LDH assays. The apoptosis induction was determined using nuclei DAPI staining and FACS assays. The main compounds of extract were identified and quantitatively analyzed using the validated HPLC method. The extract showed cytotoxic effects in various liver and breast cancer cells but had minimal effects on normal cells. It induced apoptosis on both Hep3B and SKBR3 cells in a dose-dependent manner. In addition, three sesquiterpene compounds, such as germacrone (3.25 ± 0.32 mg/g), ar-turmerone (1.12 ± 0.24 mg/g), and curcumol (0.31 ± 0.12 mg/g) were found as the main components of CSE. This is the first report on the cytotoxic effect of rhizomes against cancer cells.

甘果是一种越南药用植物,在传统和民间医学中常用于治疗各种疾病。本研究的目的是研究根茎提取物 (CSE) 的化学成分和抗增殖活性。使用 WST-1 和 LDH 测定法评估 CSE 的细胞毒性。通过核 DAPI 染色和 FACS 测定法确定细胞凋亡诱导。使用经过验证的 HPLC 方法鉴定和定量分析提取物中的主要化合物。提取物对各种肝癌和乳腺癌细胞表现出细胞毒性作用,但对正常细胞的影响最小。它以剂量依赖性方式诱导 Hep3B 和 SKBR3 细胞凋亡。此外,还发现三种倍半萜化合物,如倍半萜内酯(3.25±0.32mg/g)、莪术酮(1.12±0.24mg/g)和莪术醇(0.31±0.12mg/g),为 CSE 的主要成分。这是首次报道根茎对癌细胞的细胞毒性作用。
