To examine the expression of transforming growth factor- (TGF-) in the periapical granulation tissue and serum of patients with chronic apical periodontitis and to conduct immunohistochemical analysis so as to explore the relationship between TGF- and the degree of periapical lesions.
Periapical granulation tissues of 20 cases of chronic apical periodontitis were collected as the experimental group. Healthy gingival tissues without eruption of third molars of 5 cases were collected as the control group. Immunohistochemistry, enzyme-linked immunosorbent assay, and real-time PCR (RT-PCR) were utilized to determine the expression of TGF- mRNA and protein, and the difference in the expression of TGF- was compared between groups. In the experimental group, oral CBCT was taken to measure the periapical bone resorption area. Spearman's correlation method was applied to analyze the correlation between TGF- protein and gene expression levels and periapical bone resorption area.
Immunohistochemistry and enzyme-linked immunosorbent assay demonstrated that the expression of TGF- protein in chronic apical periodontitis tissue and serum was higher than that in the controls ( < 0.05). RT-PCR revealed that the expression of TGF- mRNA was higher in chronic apical periodontitis tissue than that of the controls ( < 0.05). Spearman's correlation analysis showed that in the experimental group, the mRNA expression of TGF- was positively correlated with the periapical bone resorption area ( < 0.01), and the protein expression level was not correlated with the periapical bone resorption area ( < 0.05).
The increased expression of TGF- in the periapical granulation tissue and serum of patients with chronic apical periodontitis has a certain correlation with the progression of periapical periodontitis. The correlation between TGF- at the mRNA level and the degree of early stage disease as well as the high expression of TGF- in inflammatory cells in immunohistochemistry have confirmed that TGF- promotes bone resorption in early periapical periodontitis, and its mechanism of action deserves further investigation.
检测转化生长因子-(TGF-)在慢性根尖周炎患者根尖肉芽组织和血清中的表达,并进行免疫组织化学分析,探讨 TGF-与根尖病变程度的关系。
收集 20 例慢性根尖周炎患者的根尖肉芽组织作为实验组,收集 5 例未萌出第三磨牙的健康牙龈组织作为对照组。采用免疫组织化学、酶联免疫吸附试验和实时 PCR(RT-PCR)检测 TGF- mRNA 和蛋白的表达,并比较两组 TGF-的表达差异。实验组拍摄口腔锥形束 CT 测量根尖骨吸收面积,采用 Spearman 相关分析法分析 TGF-蛋白与基因表达水平与根尖骨吸收面积的相关性。
免疫组织化学和酶联免疫吸附试验显示,慢性根尖周炎组织和血清中 TGF-蛋白的表达高于对照组(<0.05)。RT-PCR 显示,慢性根尖周炎组织中 TGF- mRNA 的表达高于对照组(<0.05)。Spearman 相关分析显示,在实验组中,TGF- mRNA 的表达与根尖骨吸收面积呈正相关(<0.01),蛋白表达水平与根尖骨吸收面积无相关性(<0.05)。
慢性根尖周炎患者根尖肉芽组织和血清中 TGF-的表达增加与根尖周炎的进展有一定的相关性。TGF-在 mRNA 水平上与疾病早期的相关性以及免疫组织化学中炎症细胞中 TGF-的高表达证实,TGF-促进早期根尖周炎的骨吸收,其作用机制值得进一步研究。
