Abstract

Experimental type 1 diabetes mellitus (T1DM) was induced in rats by daily intraperitoneal injections of alloxan in a dose of 90 mg/kg for 4 days. For verification of insulin resistance, insulin tolerance test was performed in 2 weeks and the glucose utilization rate constant (K) was calculated. The rats demonstrated the main symptoms of T1DM: hypoinsulinemia, hyperglycemia, ketonemia, glucosuria, ketonuria, polydipsia, polyphagia, weight loss, and insulin resistance, as evidenced by a decrease in K. The serum content of free fatty acids and triacylglycerols significantly increased. The content of triacylglycerols increased in skeletal muscles and decreased in the liver. A negative linear correlation was found between K and triacylglycerol content in muscles. Thus, the development of insulin resistance in experimental T1DM in rats is associated with accumulation of triacylglycerols in skeletal muscles.

实验性 1 型糖尿病(T1DM)通过每天腹腔注射 90mg/kg 的链脲佐菌素连续 4 天诱导大鼠产生。为了验证胰岛素抵抗,在 2 周时进行胰岛素耐量试验,并计算葡萄糖利用率常数(K)。大鼠表现出 T1DM 的主要症状:低胰岛素血症、高血糖、酮血症、糖尿、酮尿、多饮、多食、体重减轻和胰岛素抵抗,表现为 K 值降低。血清游离脂肪酸和三酰甘油含量显著增加。骨骼肌中的三酰甘油含量增加,而肝脏中的三酰甘油含量减少。K 值与肌肉中三酰甘油含量呈负线性相关。因此,大鼠实验性 T1DM 中胰岛素抵抗的发展与骨骼肌中三酰甘油的积累有关。
