This study aimed to evaluate the inflammatory response, hyperpolarization-activated cyclic nucleotide-gated 2 (HCN2), and voltage-gated potassium (Kv) 9.1 channel expression in rats with paclitaxel-induced neuropathic pain-like behavior.
Sixteen male Sprague Dawley rats were divided equally into two groups: control and paclitaxel-induced pain (PTX). The attachment loss and inflammatory cell infiltrate levels were analyzed histometrically and immunohistochemically. The gene expression of HCN2 and KCNS1 was analyzed by qPCR in the brain and gingival tissues.
The attachment loss and prominent infiltration of inflammatory cells were significantly higher in the PTX group than in the control groups. In gingival tissues; the expression levels of HCN2 (p = 0,0011) were significantly higher and KCNS1 (p = 0,0003) were significantly lower in the PTX group than in the control groups.
Increased nociceptive sensitivity, may play a role in periodontal inflammation. KCNS1 may decrease and HCN2 expression may increase in periodontium in permanent chronic pain states. The results of the present study may be helpful in developing new approaches to alleviate pain and maintain periodontal health in patients suffering from orofacial pain.
本研究旨在评估紫杉醇诱导的神经病理性疼痛样行为大鼠的炎症反应、超极化激活环核苷酸门控 2(HCN2)和电压门控钾(Kv)9.1 通道表达。
将 16 只雄性 Sprague Dawley 大鼠平均分为两组:对照组和紫杉醇诱导疼痛组(PTX)。通过组织学和免疫组织化学分析附着丧失和炎症细胞浸润水平。通过 qPCR 分析大脑和牙龈组织中 HCN2 和 KCNS1 的基因表达。
PTX 组的附着丧失和炎症细胞浸润明显高于对照组。在牙龈组织中,PTX 组的 HCN2(p=0.0011)表达水平明显高于对照组,KCNS1(p=0.0003)表达水平明显低于对照组。
疼痛敏感性增加可能在牙周炎症中起作用。在永久性慢性疼痛状态下,牙周组织中 KCNS1 表达减少,HCN2 表达增加。本研究的结果可能有助于开发减轻疼痛和维持牙周健康的新方法,以缓解口腔颌面部疼痛患者的疼痛。
