Abstract

We have developed an efficient protocol for the construction of polycyclic dihydrobenzofuran spirooxindole scaffolds via base promoted cascade annulation of Morita-Baylis-Hillman (MBH) carbonates of isatins with -hydroxychalcones or -hydroxy-β-nitrostyrenes. The complex polycyclic compounds were conveniently synthesized in satisfactory yields and with high diastereoselectivity. This protocol provides a swift and convenient approach for the assembly of diverse highly functionalized dihydrobenzofuran spirooxindoles and also features broad substrate scope, high molecular convergence, and excellent atomic economy.

我们已经开发出一种高效的方法,通过碱促进异吲哚酮的Morita-Baylis-Hillman(MBH)碳酸酯与α-羟基查耳酮或α-羟基-β-硝基苯乙烯的串联环化反应来构建多环二氢苯并呋喃螺氧化吲哚支架。这些复杂的多环化合物能够以令人满意的产率和高非对映选择性方便地合成出来。该方法为组装各种高度官能化的二氢苯并呋喃螺氧化吲哚提供了一种快速便捷的途径,并且具有底物范围广、分子收敛性高和原子经济性优异的特点。
