We compared four combinations of nasopharyngeal swabs and transport media for their ability to transfer and recover viruses under different storage conditions.
Each swab was immersed in culture supernatants of influenza A virus (IAV), respiratory syncytial virus, and adenovirus, placed in transport medium, and stored at -20℃, +4℃, +20 to 25℃, and +37℃ for 5 days. On each day, virus culture and real-time PCR were performed for each virus.
All samples under different storage conditions showed positive results up to 5 days using both virus culture and real-time PCR. Real-time PCR showed that samples stored at -20℃, 4℃, and 20 - 25℃ were within two cycle thresholds (Cts) up to 5 days, but IAV at 37℃ showed that viral titer decreased after 3 days.
Our results indicate that these swab and transport media maintained the stability of the above viruses for 5 days at room temperature, refrigerated, and frozen storage conditions.
我们比较了鼻咽拭子和转运培养基的四种组合在不同储存条件下转移和回收病毒的能力。
将每个拭子浸入甲型流感病毒(IAV)、呼吸道合胞病毒和腺病毒的培养上清液中,置于转运培养基中,并分别在-20℃、+4℃、+20至25℃和+37℃下储存5天。每天对每种病毒进行病毒培养和实时PCR检测。
在不同储存条件下,所有样本在5天内使用病毒培养和实时PCR检测均呈阳性结果。实时PCR显示,储存在-20℃、4℃和20 - 25℃的样本在5天内的循环阈值(Ct值)相差两个以内,但37℃下的IAV在3天后病毒滴度下降。
我们的结果表明,这些拭子和转运培养基在室温、冷藏和冷冻储存条件下可使上述病毒的稳定性维持5天。
