The gut microbiome (GM) of rheumatic arthritis (RA) patients is often altered in composition and function. Moreover, methotrexate (MTX), one of the most frequently used disease-modifying antirheumatic drugs, is known to negatively affect GM composition. The modulation of immune system activity is one of the therapeutic benefits of probiotics. The aim of the current investigation was to determine the impact of MTX therapy combined with one of the Lactobacillus strains, LS/07 (LB), on adjuvant arthritis (AA) in rats. Methods focused on biometric and inflammatory parameters in AA, particularly on plasmatic levels of IL-17A, MMP-9, and MCP-1, and the activities of gamma-glutamyl transferase in the spleen and joints were applied. Enhancing the effect of MTX, LB positively influenced all biometric and inflammatory parameters. The findings of the present study may be of help in proposing novel therapeutic strategies for RA patients.
类风湿关节炎(RA)患者的肠道微生物组(GM)在组成和功能上常常发生改变。此外,甲氨蝶呤(MTX)是最常使用的疾病修饰抗风湿药物之一,已知其会对 GM 组成产生负面影响。益生菌的治疗益处之一是调节免疫系统活性。本研究旨在确定 MTX 联合其中一种乳杆菌菌株 LS/07(LB)治疗佐剂关节炎(AA)在大鼠中的影响。方法侧重于 AA 的生物计量和炎症参数,特别是血浆中 IL-17A、MMP-9 和 MCP-1 的水平,以及脾脏和关节中γ-谷氨酰转移酶的活性。LB 增强了 MTX 的效果,对所有生物计量和炎症参数均产生了积极影响。本研究的结果可能有助于为 RA 患者提出新的治疗策略。
