Abstract

OBJECTIVE

To observe the effect of "lingguibafa" moxibustion at "opening" time on the level of superoxide dismutase(SOD), malondialdehyde(MDA), the protein expressions of B-cell lymphoma-2 (Bcl-2) and Bcl-2 associated X protein (Bax) in aging model rats.

METHODS

SD rats were randomly divided into blank control, model and lingguibafa groups, with 8 rats in each group. The aging model was established by daily intraperitoneal injection of D-galactose (500 mg/kg) for 42 consecutive days. After successful modeling, moxibustion intervention was applied at"lingguibafa" acupoints, 3 moxa-cone for each acupoint, once a day for 28 consecutive days. The contents of SOD and MDA in serum were detected by ELISA. Morphological changes of testicular tissue and the number of Leydig cells were observed after HE staining. Apoptosis rate of testicular cells was detected by TUNEL staining. The protein expression levels of Bcl-2 and Bax in testis were detected by Western blot.

RESULTS

Compared with the blank control group, the SOD content in the serum, the number of testicular Leydig cells, the expression level of Bcl-2 protein in testis were significantly decreased (<0.01) in the model group, while the MDA content in the serum, the apoptosis rate of testicular cells and the expression level of Bax protein in testis were significantly increased (<0.01). Compared with the model group, the SOD content in serum, the number of testicular Leydig cells, the Bcl-2 protein expression in testis were significantly increased (<0.01), while the MDA content in serum, the apoptosis rate of testicular cells, the expression level of Bax protein in testis were significantly decreased(<0.01) in the lingguibafa group.

CONCLUSION

Moxibustion on acupoints at "opening" time can delay testicular aging, and the mechanism may be related to balancing the metabolism of free radicals, reducing oxidative damage, and inhibiting the apoptosis of testicular cells.

目的

观察“灵龟八法”开穴艾灸对衰老模型大鼠超氧化物歧化酶(SOD)、丙二醛(MDA)水平及睾丸组织中B细胞淋巴瘤-2(Bcl-2)和Bcl-2相关X蛋白(Bax)蛋白表达的影响。

方法

将SD大鼠随机分为空白对照组、模型组和灵龟八法组,每组8只。连续42天每日腹腔注射D-半乳糖(500 mg/kg)建立衰老模型。造模成功后,于“灵龟八法”穴位进行艾灸干预,每穴3壮,每天1次,连续28天。采用酶联免疫吸附测定法(ELISA)检测血清中SOD和MDA含量。苏木精-伊红(HE)染色后观察睾丸组织形态变化及睾丸间质细胞数量。采用脱氧核糖核苷酸末端转移酶介导的缺口末端标记法(TUNEL)检测睾丸细胞凋亡率。采用蛋白质免疫印迹法(Western blot)检测睾丸中Bcl-2和Bax蛋白表达水平。

结果

与空白对照组比较,模型组血清中SOD含量、睾丸间质细胞数量、睾丸组织中Bcl-2蛋白表达水平显著降低(<0.01),而血清中MDA含量、睾丸细胞凋亡率、睾丸组织中Bax蛋白表达水平显著升高(<0.01)。与模型组比较,灵龟八法组血清中SOD含量、睾丸间质细胞数量、睾丸组织中Bcl-2蛋白表达水平显著升高(<0.01),而血清中MDA含量、睾丸细胞凋亡率、睾丸组织中Bax蛋白表达水平显著降低(<0.01)。

结论

“灵龟八法”开穴艾灸可延缓睾丸衰老,其机制可能与平衡自由基代谢、减轻氧化损伤、抑制睾丸细胞凋亡有关。
