To analyze the expression and correlation of microRNA-195 (miR-195), and calreticulin in diffuse large B-cell lymphoma (DLBCL).
From April 2020 to April 2021, 80 DLBCL patients with complete data archived by the Pathology Department of Handan First Hospital and The Second Hospital of Hebei Medical University were selected as the study group, and 70 patients with reactive lymph node hyperplasia were selected as the control group. The expressions of and were detected by real-time fluorescence quantitative PCR, and the expression of calreticulin was detected by Western blot. Pearson correlation was used to analyze the correlation between , , calreticulin and DLBCL, and ROC curve was used to analyze the predictive value of , and calreticulin for DLBCL.
Compared with the control group, the expression of decreased but and calreticulin increased in the study group (<0.001). The expression levels of , and calreticulin were not related to sex, age, primary site and B symptoms of patients with DLBCL, but related to immunophenotype, Ann Arbor stage, lactate dehydrogenase, IPI score, nodule involvement and Ki-67 index. The expression of decreased and the expression of and calreticulin increased in DLBCL paitents with non-germinal center source, Ann Arbor stage III-IV, lactate dehydrogenase > 245 U/L, IPI score 3-5, nodule involvement≥2 and Ki-67 index≥75% (<0.05). Pearson correlation analysis showed that and were negatively correlated (=-0.536, =0.001), and calreticulin were negatively correlated (=-0.545, =0.001), while and calreticulin were positively correlated (=0.523, =0.001). ROC curve showed that compared with the single diagnosis of , and calreticulin, the combination of the three items had higher predictive value for DLBCL (<0.001).
The expression of decreases and the expression of and calreticulin increase in patients with DLBCL. Along with the increase of disease stage and IPI score, the decrease of and the increase of and calreticulin aggravate gradually. The three items may participate in the occurrence and progress of DLBCL.
分析微小RNA-195（miR-195）及钙网蛋白在弥漫性大B细胞淋巴瘤（DLBCL）中的表达及相关性。
选取2020年4月至2021年4月在邯郸市中心医院及河北医科大学第二医院病理科存档的80例资料完整的DLBCL患者作为研究组，选取70例反应性淋巴结增生患者作为对照组。采用实时荧光定量PCR检测miR-195及钙网蛋白的表达，采用蛋白质免疫印迹法检测钙网蛋白的表达。采用Pearson相关性分析miR-195、钙网蛋白与DLBCL的相关性，采用ROC曲线分析miR-195、钙网蛋白对DLBCL的预测价值。
与对照组相比，研究组miR-195表达降低，钙网蛋白表达升高（P<0.001）。miR-195、钙网蛋白的表达水平与DLBCL患者的性别、年龄、原发部位及B症状无关，但与免疫表型、Ann Arbor分期、乳酸脱氢酶、国际预后指数（IPI）评分、结外受累及Ki-67指数有关。非生发中心来源、Ann Arbor分期Ⅲ-Ⅳ期、乳酸脱氢酶>245 U/L、IPI评分3-5分、结外受累≥2处及Ki-67指数≥75%的DLBCL患者miR-195表达降低，钙网蛋白表达升高（P<0.05）。Pearson相关性分析显示，miR-195与钙网蛋白呈负相关（r=-0.536，P=0.001），miR-195与钙网蛋白呈负相关（r=-0.545，P=0.001），而钙网蛋白与钙网蛋白呈正相关（r=0.523，P=0.001）。ROC曲线显示，与单独诊断miR-195、钙网蛋白相比，三者联合对DLBCL具有更高的预测价值（P<0.001）。
DLBCL患者miR-195表达降低，钙网蛋白表达升高。随着疾病分期及IPI评分的增加，miR-195降低及钙网蛋白升高逐渐加重。三者可能参与DLBCL的发生及进展。
