To investigate the effect of shikonin (SKN) on proliferation, apoptosis, migration and angiogenesis of hemangioma endothelial cell (HemEC).
CCK-8 and EdU assays were used to detect the effect of SKN on proliferation of HemEC. The effect of SKN on apoptosis of HemEC was detected by flow cytometry. Wound healing assay was used to detect the effect of SKN on migration ability of HemEC. The effect of SKN on angiogenesis ability of HemEC was detected by tube formation assay. SPSS 22.0 software package was used for statistical analysis of the data.
SKN inhibited proliferation (P<0.001) and promoted apoptosis (P<0.001) of HemEC in a concentration-dependent manner. In additon, SKN inhibited HemEC migration(P<0.01) and angiogenesis(P<0.001).
SKN can inhibit proliferation, migration, angiogenesis and promote apoptosis of HemEC.
探讨紫草素(SKN)对血管瘤内皮细胞(HemEC)增殖、凋亡、迁移及血管生成的影响。
采用CCK-8和EdU检测法检测SKN对HemEC增殖的影响。通过流式细胞术检测SKN对HemEC凋亡的影响。采用伤口愈合实验检测SKN对HemEC迁移能力的影响。通过管腔形成实验检测SKN对HemEC血管生成能力的影响。使用SPSS 22.0软件包对数据进行统计分析。
SKN以浓度依赖性方式抑制HemEC的增殖(P<0.001)并促进其凋亡(P<0.001)。此外,SKN抑制HemEC的迁移(P<0.01)和血管生成(P<0.001)。
